Objective: To study the HSP27protein expression of ovarian cancer cells and tissuesand the relationship between HSP27and the DDP resistance and prognosis of oophoromapatients. To investigate the mechanism of HSP27in drug resistance of cisplatin inoophoroma cells.Methods: Western-blot and Real-time RT-PCR methods was used to detect theexpression of HSP27gene in ovarian cancer cells. Immunohistochemical methods was usedto detect the expression of HSP27protein in oophoroma tissues from101cases ofoophoroma patients and10cases of normal and benigh ovarian tissues. SiRNA weretransfected into C13K, detected the drug sensitivity by CCK-8and flow cytometry.Immunofluorescence was used to detect the expression of P21.Results: The expression of HSP27protein in C13K cells were significantly higher thanthose in the other5cell lines (p<0.05). The IC50to cisplatin of C13K was obviously higherthan the other cell lines. HSP27expression was significantly higher in oophoroma tissuethan normal ovaries. The patients in the high expression group of HSP27had a lowermedian PFS than the low expression group. Transfection the siRNA of HSP27into C13Kcells, result that C13K cells apoptosis rate significantly increased than NC cells, the HSP27and P-Akt protein levels were reduce. Immunofluorescence results showed that P21transferred from cytolymph to nucleus in si group and the LY294002group.Conclusion: HSP27gene expressions in C13K cells can affect the sensitivity tocisplatin. In C13K cells, inhibit the expression of HSP27, resulted in depressing thephosphorylation of Akt, promoted the P21transferred from cytolymph to nucleus, increased the apoptosis of oophoroma cells.
