Objective: Bisphenol A (BPA) is a kind of environmental endocrine disrupters, itexists in the food and beverage packaging, adhesive, construction materials, electroniccomponents, and in paper coating. Prior researches suggest that BPA may haveadverse effects on the reproductive development, immune system, metabolic system,brain development and psychological health. And some studies have shown that BPAhas adverse effects on male reproductive health, but under the environmentalexposure level whether damage effect exist is unclear. The aims of the present studyare to observe the effects of bisphenol A on the reproductive system of adult maleWistar rats by long-term treatment of BPA about environmental exposure dose, and toexplore the mechanism of bisphenol A on reproductive function.This study willprovid evidence of the effects of bisphenol A (BPA) on male reproductive healthresearch for further research.Methods:40adult male Wistar rats were randomly divided into4groups according tothe dose of BPA: control group (C)(0μg/kg.bw/day), high dose group (B250)(250μg/kg.bw/day), middle dose group (B50)(50μg/kg.bw/day), low dose group (B10)(10μg/kg.bw/day). Different doses of bisphenol A (BPA) were dissolved in the samevolume of corn oil, to get different concentrations of mixture. Take0.3ml mixture,blended with5g normal rat feed which got the feed containing different dose ofbisphenol A. The control group used0.3ml corn oil mixed with the feed directly.During breeding animals, at8:00AM to give5g mixed rat feed,5:00PM to give rightamount of rats normal feed. The experiment lasted for35weeks. At the end of experiment, all rats were sacrificed. The liver, kidney, heart, testis and epididymistissue were separated and weighted. Epididymis tissue were prepared to measure therate of abnormal sperm. The mRNA expressions of suicide related factor (Fas/FasL),aspartic acid specific enzyme of cysteine proteinase3(Caspase3), G protein coupledreceptor (GPER) in testis of rats were quantified by Real-time PCR.Results:1. No significant difference was found between control group and BPAgroups on body weight, coefficient of liver, coefficient of kidney, coefficient of heart,coefficient of testis, coefficient of epididymis.(P>0.05).2. Sperm abnormality rate in highest dose BPA group (BPA250) was high than controlgroup(P<0.05).3. The level of FasL and Caspase3mRNA in rat testis were increased in comparedwith that of control group(P<0.05).Conclusion: Under this experiment conditions, long term exposure BPA to adult malerats on250μg/kg.bw/day, we observed some effects, but not observed damage effectson rats at environmental dose of BPA.
