Objective: This research aim to study the specifi effect and mechanism of acupuncture“He-Mu” combination acupoint to prevent and treat acute gastritis by using infrared thermalimaging and molecular biology technology in guidance of TCM acupuncture theory, and discussthe effect and synergy mechanism of acupuncture “He-Mu” combination acupoint to prevent andtreat acute gastritis, then provide ideas and basis for the research of acupoint and synergisticeffect.Methods: The60SD rats (180-220g, male) were randomly divided into six groups applyingthe method of random number table generated by computer as follows, blank control group, grabgroup, model group,“Zhongwan”(RN12) group,“Housanli”(ST36)group, and “He-Mucombination acupoints” group(“Zhongwan” matching “Housanli”), each group has10rats. Adoptpotent liquor to duplicate rat model of acute gastritis. Each treatment group in accordance withthe relevant requirements to take corresponding acupoints, and adopt acupuncture therapy oftwirling reinforcing reducing (twist amplitude of180°, the frequency of60~80times/min),1time per day and20minutes per time. Acupuncture continuously6days before duplicating ratmodel, and acupuncture continuously3days after duplicating rat model. We take thermal infraredspectrum of gastric surface projection area before and after acupuncture every day by infraredimaging devices, extract and analyse the temperature, then detect the cox-2mRNA expression ofgastric tissue and the hypothalamus with real-time fluorescence quantitative polymerase chainreaction (qRT-PCR) technology after the acupuncture treatment.Results:1. Macroscopic observation results of gastric tissue of all groups: rats’ gastric mucosal hadno obvious changes among blank control and grab group. The rats’ gastric mucosal in modelgroup were incomplete, and there has obvious hyperemia and edema, erosion, bleeding andobvious point sheet on mucosal surface. There were different degrees of improvement on rats’gastric mucosal of “Zhongwan” group,“Housanli” group, and “He-Mu combination acupoints”group, especially rats in the “He-Mu combination acupoints” group showed obvious effect.2. Analysis of thermal infrared spectrum of all groups(1) The thermal infrared spectrum has no obvious abnormalities on rats’ gastric body surfaceprojection area of blank control and grab group; The thermal infrared spectrum showed abnormallow temperature area on the rats’ gastric body surface projection area of model group; the lowtemperature area on rats’ gastric body surface projection area that showed in thermal infrared spectrum improved to varying degrees in three acupuncture groups, indicated that lowtemperature area decreased.(2) During the experimental period, the temperature has no obvious change on rats’ gastricbody surface projection area of blank control and grab group; After modeling, the temperaturedecreased significantly on7,8,9thday in model group (P<0.01), suggested that alcohol bygavage had a obvious effect on the temperature on gastric surface projection area; On the7thday,the temperature decreased significantly on gastric surface projection area of three acupuncturegroups (P<0.01); On the7and8thday, the temperature had different degree of recovery(P<0.01),especially in“He-Mu combination acupoints” group.The temperature of the gastric surface projection area increased obviously on the8and9thday in “He-Mu combination acupoints” group when compared with model group, thedifference was significant (P<0.01); The temperature of the gastric surface projection areaincreased obviously on the9thday in “Zhongwan” group and “Housanli” group, the differencewas significant (P<0.01), indicated that acupuncture “Zhongwan”,“Housanli” and “He-Mucombination acupoints” can increase the temperature of gastric body surface projection area. Thetemperature of the gastric surface projection area increased bigger on the8thday in “He-Muacupoints” group when compared with “Zhongwan” and “Housanli”group, the difference wassignificant(P<0.01); indicated that acupuncture “He-Mu combination acupoints” had a moresignificant effect and shorter schedule on increasing temperature of gastric body surfaceprojection area, showing a strong specificity.3. Detection and analysis of cox-2mRNA expression on rats’ gastric tissue andhypothalamus:(1) The expression of cox-2mRNA on gastric tissue increased in grab group when comparedwith blank control group, the difference was significant(P<0.05); The expression of cox-2mRNA in gastric tissue increased in model group when compared with grab group, the differencewas significant(P<0.01); The expression of cox-2mRNA in gastric tissue decreased in threeacupuncture groups when compared with model group, the difference was significant(P<0.01);When compared within the three acupuncture groups, there were no significant difference(P>0.05).(2) The expression of cox-2mRNA in hypothalamus increased in grab group when comparedwith blank control group, the difference was significan(tP<0.05); The expression of cox-2mRNAin hypothalamus increased in model group when compared with grab group, the difference wassignificant(P<0.01); The expression of cox-2mRNA in hypothalamus decreased in three acupuncture groups when compared with model group, the difference was significant(P<0.01);within three acupuncture groups, the expression of cox-2mRNA in hypothalamus was lowestwhen compared with “Housanli” group, the difference was very significant (P<0.01), whencompared with “Zhongwan” group, the difference was significant (P<0.05).4. Correlation analysis of cox-2mRNA expression on rats’ gastric tissue and hypothalamus:The temperature of the gastric surface projection area decreased obviously in model groupwhen compared with grab group (P<0.01), however, the expression of cox-2mRNA in gastrictissue and hypothalamus increased Significantly(P<0.01). The temperature of the gastric surfaceprojection area decreased obviously in three acupuncture groups when compared with modelgroup (P<0.01), while the expression of cox-2mRNA in gastric tissue and hypothalamusdecreased Significantly (P<0.01). And the rate of reduce of cox-2mRNA in gastric tissue andhypothalamus in “He-Mu combination acupoints” group was larger than that of “Zhongwan” and“Housanli” group(P <0.01).Conclusion:(1) Potent liquor can damage obviously to gastric mucosa of rats, thermal infrared spectrumof gastric surface projection area showed abnormal low temperature area, both “He-Mucombination acupoints” g and single point can improve the injury of gastric mucosa, recover thetemperature of gastric area, the effect of acupoints was better than single point.(2) The mechanism of acupuncture treatment and prevention of alcoholic gastritis may berelated with the inhibition of cox-2mRNA expression of gastric tissue and hypothalamus; Inreducing the cox-2mRNA expression on hypothalamus, the effect of acupoints (“He-Mu”combination acupoint) was better than the single point (“Zhongwan” and “Housanli”), the effectwas specific.(3) Temperature changes of gastric zone related reversly with the expression of cox-2mRNAon rats’ gastric tissue and hypothalamus, while the expression of cox-2mRNA on hypothalamusand gastric tissue may had some kind of inner link, the specific mechanism and methods needfurther research.
