Methods:24SD rats were randomly divided into4groups,production of sciatic nerve injury model in the greater sciatic foramen. Group A: the sham operation group, only revealed nerve; group B: denervation group, the nerve complete disconnection does not match; group C: nerve section anastomosis group, the nerve complete disconnection part anastomosis; group D: nerve anastomosis group, the nerve complete disconnection completely anastomosis. After4,8weeks, Determination of the recoveryof neurological function by the sciatic nerve function index (SFI), nerve conduction velocity (NCV), nerve anastomosis through rate (nerve fiber count), the situation of muscle atrophy (gastrocnemius muscle wet weight ratio, muscle fiber cross-sectional area).Incision of lateral soleus muscle samples at middle10mg for real-time fluorescence quantitative PCR detection of miRNA-206specific expression.Results: The condition of different healing In compliance with molding of the sciatic nerve function index of rat sciatic nerve after injury with fracture (SFI), nerve conduction velocity (NCV)determination, nerve anastomosis through rate, muscle atrophy; denervation (denervation group) muscle specific miRNA-206expression in4weeks,8weeks were2.09±0.17,1.83±0.07, no significant difference; reinnervation (nerve sectionanastomosis group, nerve anastomosis group) muscle specific miRNA-206expressionat4weeks were2.45±0.23,3.08±0.41,8weeks respectively2.58±0.15,3.20±0.20, were higher than the denervation group, there is significant significant differences; reinnervation of nerve anastomosis group, nerve anastomosis group relative expression ofmuscle specific miRNA-206in4weeks,8weeks, there was significant difference.Conclusion:1the sciatic nerve function index (SFI), nerve conduction velocity (NCV)determination, nerve fiber count, muscle atrophy can be used as indicators of response function in neural experiment;2.Real-time fluorescent quantitative PCR Application of detection of muscle specific miRNA-206expression, early, accurate, and reliable todetect the nerve recovery, can understand the neural fiber regeneration.
