Research Of Clinical Application Of IFN-γ Release Assays And Related Cytokines Expression In Mycobacterium Tuberculosis Infected Cases

Objective: To evaluate clinical application of IFN-γ release assays in detecting activetuberculosis, inactive tuberculosis, latent tuberculosis infection cases and other diseasessufferings. And to detect the expression levels of TGF-β1, IL-17, IL-21, IL-10and IL-23inactive tuberculosis, inactive tuberculosis, latent tuberculosis infection cases and healthysubjects for understanding the function of these cytokines in these diseases.Methods:489cases were collected from January1to April30,2014in affiliatedhospital of Zunyi Medical College, including active tuberculosis, inactive tuberculosis,latent tuberculosis infection cases and other diseases patients. IGRA detection wasperformed, and meanwhile TGF-β1, IL-17, IL-21, IL-10and IL-23expression levels weretested by ELISA technology between tuberculosis infections and health controls.Results: In489samples,212were active tuberculosis,30were non-active tuberculosis,91latent tuberculosis and156other diseases patients. Active TB patients were divided intobacterium-positive and bacterium-negative groups,11cases in14bacterium-positive caseswere positive for IGRA assay, the positive rate was78.6%(11/14),157in198bacterium-negative cases were positive for IGRA assay,39cases negative, and2casesuncertainty, the positive rate was79.3%(157/198).24in30inactive tuberculosis caseswere positive,6were negative, positive rate80.0%(24/30). All91latent tuberculosiscases’IGRAwere positive.150in156other diseases patients’IGRAwere negative,6wereuncertain.TGF-β1, IL-17, IL-21, IL-10and IL-23cytokines increased in active tuberculosis,inactive tuberculosis and latent tuberculosis patients might have a certain clinicalapplication value.Conclusion: IGRA plays an important role in diagnosing of Mycobacterium tuberculosislatent infection, but comprehensive analyses are essential in case of abnormal immunestatuses. In this study, the expression of Cytokines TGF-β1, IL-17, IL-21, IL-10and IL-23in active tuberculosis, inactive tuberculosis, and latent tuberculosis patients inperipheral blood have some clinical significance.
