Purpose: Recent studies suggested that adiponectin (APN) wasassociated with several retinal diseases. We studied the expression of APNand its receptors (AdipoRs) in the retina of human and a mouse model oftype1diabetes mellitus (T1DM).Methods:Human eyeball specimens were obtained from theChongqing Eye Bank. eNOS-knockout (eNOS-/-) mice were randomlydivided into T1DM group and control group. T1DM model was induced byintraperitoneal injection of streptozotocin. To locate the AdipoRs in theretina, immunofluorescence was performed. Total APN protein and RNAwere extracted from Retina and retinal pigment epithelium (RPE)-choroid,and the APN protein was detected by ELISA. The mRNA and protein ofAdipoRs in the retina was detected respectively by qRT-PCR and westernblotting. Unpaired Student’s t-test was used to assess the significancebetween the T1DM and the control groups, with p<0.05was regarded asstatistically significant.Results: APN, AdipoR1and AdipoR2were identified in retina and theRPE-choroid of human and mice. AdipoR1was found in internal limiting membrane and photoreceptor outer segments both in human and mouseretina, whereas no noticeable AdipoR2expression was seen in the retinalfrozen sections of human and mouse eyes. Compared to the control group,expressions of APN and AdipoR1in the retina were elevated in the T1DMgroup, but expressions of AdipoR2remained unchanged.Conclusions: Expressing of APN, AdipoR1and AdipoR2in the retinaand choroid of human and mouse suggests APN system may play a role inmaintainning retinal function. Retinal APN and AdipoR1protein levelswere elevated in T1DM mice implying APN-AdipoR1axis may beactivated in diabetic retina, whereas it appeared that AdipoR2played aminor role in this pathologic process.
