Objective:The bioprosthetic heart valves advantage is excellent hemodynamic, Less damage to blood composition, don’t need to long-term anticoagulation and less complications. But it easy to deteriorate. The bioprosthetic heart valves most used in older patients or to need preserve fertility in female patients. Calcification is a major reason of deterioration of Bioprosthetic heart valves in the body. This experiment using decellularization and surface heparin treatment, evaluation of bioprosthetic heart valves calcification and inflammatory cells invasion situation, explore new ways to prevent bioprosthetic heart valves calcification.Methods:Take fresh porcine heart valve, divided into A, B, C and D groups. Group A is fresh control group, group B used decellularization by detergent extraction and enzymatic(DEE) method, group C used by decellularization and glutaraldehyde cross-linking, group D is decellularization and glutaraldehyde cross-linking and surface heparin treatment. Each groups observe by the microscopic examination and scanning electron microscope, implanted into New Zealand white rabbit and take out after60days. Microscopic examination observed calcification and inflammatory cells invasion, microcosmic structure observed by scanning electron microscopy(SEM), used methyl thymol blue colorimetry(MTB) to examine the contents of calcium.Results:Microscopic examination observation, inflammatory invasion and calcium contents of Group D less than else groups. Methyl thyme phenol blue colorimetry to detect the contents of calcium, group A0.1973±0.1147mmol/mg, group B0.1134±0.0411mmol/mg, group C0.0892±0.0491mmol/mg, group D0.0193±0.0118mmol/mg. By SPSS16.0statistical software repeated anova, p<0.05, group D calcium contents is less than the other groups has statistics significance.Conclusions:The anticalcification ability of gorup D porcine heart valves which treated by decellularization and surface heparin treatment is better than the other group, and almost no inflammatory cell invasion.
