Clinical And Experimental Study On The Effect Of Lung Tumor Inhibitory Ointment Combined DC-CIK Cell In The Treatment Of Advanced Adenocarcinoma Of The Lum

Objective：To study the clinical effect of Lung Tumor inhibitory ointment CombinedDC-CIK cell in the Treatment on lung adenocarcinoma patients and experimental study ofDC-CIK and lung tumor inhibitory ointment containing serum sensitized DC-CIK cellkilling of A549cells.Methods：92cases of the selected lung adenocarcinoma patients were randomlydivided into Lung Tumor inhibitory ointment group (group A30cases), Chemotherapyand immune support group (group B30cases), DC-CIK lung tumor inhibitory ointmentgroup (group C30cases), a simple DC-CIK group (group D30cases). A group weretreated with lung tumor inhibitory ointment, B group were treated with the Chemotherapyplus immune support，C group were treated with the routine use of traditional Chinesemedicine plus culture medium reinfusion, D group were treated with culture mediumalone reinfusion. After four months, we observed the changes in the main symptoms,lesions change, survival time, KPS score in the state of life, body mass index, and the levelof serum Survivin.(This topic is a sequential subject, the author just reported C, Dgroups)In the experimental study, we collected anticoagulated peripheral bloodmononuclear cells (PBMC) from healthy donors, then induced DC and CIK cellsconventionally. The typical DC and DC pulsed by lung tumor inhibitory ointmentcontaining serum were co-cultured with CIK cells, respectively. Then the morphology ofDC was observed under an inverted microscope, the phenotypes of CIK cells weredetermined by flow cytometry, The cyto-toxicity of effector cells on A549cells in vitrowere measured by MTT assays. Result：In the clinical study,the Group showed better than the Group in improvingsymptons,improving quality of life and prolonging survival period（P＜0.05）.Theexperimental study suggests that by lung tumor inhibitory ointment containing serumsensitized cytotoxic activity of A549cells is higher than that of DC-CIK cells (P <0.05).Conclusion：Refer A, B groups reported in sequential issues：Lung tumor inhibitoryointment combined DC-CIK cell therapy can be more effective in improving clinical mainsymptoms and improving the quality of life and prolonging survival of advanced lungadenocarcinoma patients. But in narrow lesions, lung tumor inhibitory ointment combinedDC-CIK cell therapy and chemotherapy and immune support therapy is superior to lungtumor inhibitory ointment and pure DC-CIK therapy. According to the clinicalobservation，Lung tumor inhibitory ointment combined DC-CIK therapy significantlylowered the expression of Survivin which is superior to simple DC-CIK treatment. Theexperiments show that DC can enhance the killing activity of CIK A549cells by lungtumor inhibitory ointment containing serum sensitized.
