Background and Objective:Oxidative stress is an important reason of the occurrence and development ofchronic complications of type2diabetes.NF-κB plays an important role in oxidativestress reaction. NF-κB may be involved in the pathogenesis of chronic complicationsof diabetes mellitus.This study was to investigate the relationship between-94ins/delpolymorphism of NFKB1gene and diabetic microvascular complications (includingdiabetic nephropathy and diabetic retinopathy) in chinese han patients with type2diabetes.Methods:992patients with type2diabetes(T2DM) and210subjects with normal glucosetolerance(NGT) in Jiangxi Province were enrolled in this study.According to24hoursmicroalbuminuria,Urine protein quantitative, serum creatinine and endogenouscreatinine clearance,the T2DM patients were divided into three groups: withoutnephropathy group(DN-0), incipient nephropathy group(DN-1) and clinicalnephropathy group(DN-2); According to the retinopathy,,the T2DM patients werealso divided into two groups: without retinopathy group(NDR) and with retinopathygroup(DR). The genotype and allele frequencies of polymorphism-94ins/del inNFKB1gene were detected by using the Polymerase chain reaction–polyacrylamidegelelectrophoresis (PCR-PAGE) method. The data was analysized by SPSSl7.0statistical software.Results:1. There are three genotypes homozygous-insert (ins/ins), heterozygotes (ins/del),homozygous deficiency (del/del) of polymorphism in promoter region-94ins/del ofNFKB1gene in NGT group and T2DM group.There are no statistically significantdifferences in genotype and allele frequencies of polymorphism-94ins/del betweenNGT group and T2DM group(P>0.05).2.There were no statistically significant differences in genotype and allele frequencies of polymorphism-94ins/del among DN-0group,DN-1group and DN-2group (P>0.05).3.There were no statistically significant differences in genotype and allelefrequencies of polymorphism-94ins/del between NDR group and DR group(P>0.05).4.There were no tatistically significant differences in age, gender, known-duration of diabetes, BMI, FBG,2hBG, HbAIc, FCP,2hCP, TC, TG, HDL-C,LDL-C, SBP and DBP among T2DM patients with the genotype homozygous-insert(ins/ins), heterozygotes (ins/del) and homozygous deficiency (del/del). However, thelevels of FCP、2hCP in the del/del genotype group were statistically significantlylower than those in the ins/ins genotype group(P<0.05). no significant difference wasfound in others (P>0.05).Conclusions:1. Del/del genotype may be one of the risk factors of pancreatic beta cellfunction in patients with type2diabetes mellitus.2. Polymorphism-94ins/del of NFKB1gene is not related to diabeticnephropathy in Chinese Han nationality patients with type2diabetes.3. Polymorphism-94ins/del of NFKB1gene is not related to diabetic retinopathyin Chinese Han nationality patients with type2diabetes.
