Objective:To explore the association between eNOS gene G894T polymorphism and MildCognitive Impairment by studying distribution of allele frequencies of the genesegment between patients with Mild Cognitive Impairment and normal controlpopulation in Jiangxi province.Methods:Subjects were recruited in the first affiliated hospital of Nanchang Universitybetween January and August2012.116Mild cognitive impairment patients who werediagnosed by the department of Psychosomatic Medicine made up the experimentalgroup.116healthy physical examinees who matched gender, age and race withexperimental group formed the normal control group. MCI subjects had physical andneurological examinations by MoCA, ADL, GDS, HIS. Genomic DNA was extractedfrom EDTA-anticoagulated whole blood samples using BloodGen Mini Kit.eNOSgene G894T genotype was performed using polymerase chain reaction-restrictionlength polymorphism assays. Last, the data were analyzed by SPSS18.0statisticsoftware. The alleles and genotypes frequency of eNOS gene G894T were calculated,then Chi-square analysis was used to compare genotypic and allelic distributionbetween controls and cases (significance levelα=0.05, two sided test). For the former,we obtained the Hardy-Weinberg adjustment. In order to study the possible geneticeffects on cognitive performance among MCI individuals, analysis of t-testcomparisons for independent samples were used(significance levelα=0.05, two sidedtest).Results:1. The eNOS gene G894T genetypes of controls were in Hardy-Weinbergequilibrium(P﹥0.05);2. The frequency of the T allele in MCI group was higher than that in controlgroup(32.8%vs22.8%,χ~2=7.114,P<0.05);3. In MCI subjects, subjects carrying the T variant performed siginificantly worse in visual memory and abstract thinking function(P﹤0.05). There was nosignificant difference in MoCA(P﹥0.05).Conclusion:The eNOS gene G894T is likely association with MCI;and effects thecognitive performance of MCI patients.
