| Objective:Allergic asthma is a common clinical disease. The incidence of Allergic asthma is higher, and it easily changes into the chronic phase, seriously affect the patient’s physical and mental health, and bring huge burden to the society. Due to the limitations of Western medicine, Chinese medicine treatment of asthma is quite popular, especially the acupoint sticking is widely used in clinical. But current studies are more limited to confirm and clarify of the effect, or how to increase the skin (mucosal) absorption in order to improve the concentration of drugs, enhance the curative effect, etc. How to build a safe process of practice in patches drugs to treat asthma is not received due attention, especially there are few studies such as drug sticking cure asthma effect whether or not produce non-drug factors. Therefore, though drug sticking to treat asthma has a high-profile, but always has not yet done the basic research. Based on this, this study takes the different parts selection under the guidance of TCM theory, aims to explore the effect of prevention and treatment of asthma in Chinese medicine sticking point, and to further explore the relative specificity of acupoints, for choose clinical selection of acupoints much more accurate, make use of the experimental basis of acupoint sticking prevention and treatment of asthma.Method:This experiment takes56guinea pigs randomly into7groups:model group, drugs of sticking the acupuncture point group, drugs sticking "Fei Shu" group, drugs sticking "Dan Zhong ’group, drugs sticking Gb30group, aminophylline group, positive control group, normal control group, each group are eight. The guinea pigs of normal control group raised the same period of other groups; First six asthma model group with egg albumen sensitization method after feeding2weeks. Each acupoint sticking therapy from induced began to give Chinese medicine acupoint on the second day. We take the hair removal area of about3cm*2cm that sticking in of the guinea pig, fix the Chinese medicine cake in points of guinea pigs each treatment group,1time per day,2hours each time,2weeks to post7times of one treatment. Normal control group does not grant any processing, model group with physiological saline infiltration gauze piece instead of drugs. Then unified materials, observe the change of lung tissue and blood serum IL-4, IFN-gamma levels of guinea pigs among different groups, revealed the differences between Chinese medicine sticking point group and the effect of preventing and controlling asthma group, further observe the change of acupuncture point group and non acupoint group between guinea pig Na+-K+ATPase activity and the cx43mean optical density value, understanding the relative specificity of acupoints. Result:From ancient and modern literature and research,we have found that the key of allergic asthma pathogenesis is "phlegm retention fu lung", Zhongjing had put forward "phlegm drinkers, when the appropriate temperature of medicine" and this treatment principle is of great importance in healing asthma’s history. Get the following results from animal experiments.1. The guinea pig lung tissue of eosinophils, lymphocytes and macrophages infiltration degree increased obviously in the normal group (P<0.01); the lung tissue of eosinophils, lymphocytes and macrophages infiltration degree that in aminophylline group of is significantly lower than the model group (P<0.01); In drugs sticking "Fei Shu " group, the lung tissue of eosinophils, lymphocytes and macrophages infiltration degree is significantly lower than the model group (P<0.01), and no significant difference with positive control group (P>0.05); In drugs sticking "Dan Zhong "group, eosinophils, lymphocytes and macrophages infiltration degree in lung tissue is significantly lower than the model group (P<0.01), but drugs sticking "Fei Shu " group had significant difference (P<0.05); The lung tissue of eosinophils, lymphocytes and macrophages infiltration in sticking the Gb30group and the drugs sticking non acupoint group, increased obviously in the normal group (P<0.01), has no significant difference compared with model group (P>0.05).2. The serum IL-4in guinea pig express significantly increased in the model group than the normal group (P<0.01), and IFN-gamma express obviously declined compared with the normal group (P<0.01); The serum IL-4in guinea pig that in aminophylline group, the drugs sticking "Fei Shu " group and drugs sticking "Dan Zhong" groups express significantly decreased compared with the model group (P<0.01), the expression of IFN-gamma increased significantly compared with the model group (P<0.01); The expression of IL-4in serum of drugs sticking "Fei Shu " group decreased significantly compared with drugs sticking "Dan Zhong" group (P<0.01), the expression of IFN-gamma in drugs sticking "Dan Zhong" group increased significantly (P<0.01), and has no significant difference compared with positive control group (P>0.05);The expression of IL-4in serum of drugs sticking Gb30group and drugs sticking the acupuncture points group, increased significantly compared with the normal group (P<0.01), and the expression of IFN-gamma declined obviously compared with the normal group (P<0.01), has no significant difference compared with model group (P>0.05).3. The Na+-k+ATPase in "Fei Shu","Dan Zhong" local tissue after sticking with Chinese traditional medicine higher than non acupoint group, and have significant difference (p<0.05).4. In the normal control group,"Fei Shu" acupuncture points and side,"Dan Zhong" acupuncture points and side, the mean optical density value of cx43is low in the corresponding parts compared with the drug sticking group. The mean optical density value of cx43in each acupuncture group of drug sticking is significantly higher than the control group acupuncture points (all P<0.01). The average optical density value of cx43in Sticking group "Fei Shu" acupoints is significantly higher than the patches beside a group of acupoints (P<0.01); The average optical density value of cx43in control group "Fei Shu" acupoints is significantly higher than the control group acupuncture points side (P<0.01); the results of "Dan Zhong" is same as"Fei Shu".Conclusion:Guided by the treatment principles of "phlegm drinkers, when the appropriate temperature of medicine", primarily use the "white mustard seed coating method", and use the Chinese traditional medicine in sticking treatment of asthma, we can have the following conclusions:1. Take drugs sticking "Fei Shu" group, drugs sticking "Dan Zhong " group, drugs sticking "Gb30" group, drugs sticking non acupoint group intervene in guinea pig asthma model respectively, the curative effect is different. Drug sticking "Fei Shu" group has the bestcurative effect, that improved acupuncture point value is very important in drug patches. In this study the aminophylline treatment group had a similar effect.2. Take the drugs sticking "Fei Shu" group, drugs sticking "Dan Zhong" group, drugs sticking non acupoint group intervene in guinea pig asthma model respectively. The Na+-K+ATPase of change reflect that, acupuncture has a higher level of metabolism and overall ability from one side.3. Take the drugs sticking "Fei Shu" group, drugs sticking "Dan Zhong" group, drugs sticking non acupoint group intervene in guinea pig asthma model respectively. The protein cx43maybe one of the main composition of gap junction. It offers a new approach for understanding the mechanism of action of acupuncture points and preliminary clarify meridian scientifically. This experiment showed that has very important value of taking drugs sticking in acupuncture points to asthmatics, acupuncture point has relative specificity, for us to choose more accurate acupoints in clinical, make the experimental basis of us to use the acupoint sticking for prevention and treatment of asthma. |
