Objective:The purpose of our study was to investigate effects of components of KBL on the expressions of mRNA and corresponding protein for tyrosinase and MITF in B16murine melanoma cells,and then to explain the molecular basis on the the mechanism of treatment of vitiligo by components of KBL.The results established the basis on cilinical research and application in the future.Methods:B16murine melanoma cells were cultivated in vitro.The cells from the same bottle were transplanted to four bottles,then they were incubated for48hours.With5,15,25mg/1components of KBL effected on melanoma cells respectively for24hours,the real time-PCR and western blot methods detected components of KBL on mRNA transcription of TYR,MITF and the corresponding protein under different concentrations.Results:The B16murine melanoma cells were cultivated in vitro with components of KBL for24hours.we found that components of KBL had different degree up-regulation to mRNA transcription of TYR and its protein expression(P<0.05),and it is especially related to concentrat-ions(P<0.01).While the mRNA of MITF and its protein level had been increased(P<0.05),and it’s especially related to concentrations(P<0.01).Conclusions:The components of KBL promoting melanogenesis is related to raising TYR expression.Components of KBL on promoting melanogenesis is probable to directly regulate gene transcription and expression of the nuclear transcription factor MITF,which in turn mediated activation of tyrosinase family members’ promoter.The result may be served as the fundamental mechanism of the promotion of melanogenesis with components of KBL.
