Objectives: The objectives of the this study is to check the effects on the growth ofcells, tyrosinase activity and melanogenesis after treating A375human melanomacells by using various doses of BCG-PSN, thymosin.Methods: Dealing with A375human melanoma cells with different concentration ofBCG-PSN and thymosin;To observe the growth of A375human melanoma cells byMTT, testing the changes of tyrosinase activity with dioxypenylalanineoxidation;detecting the content of melanin with the law of spallation of NaOH.Results:1.If the concentration of BCG is between0.1ug/ml and10ug/ml,it shows agrowing tendency of the amount of A375human melanoma cells as the concentrationof Thymosin is raised. On the contrary,Thymosin dose not have a strong influence oncell proliferation between the concentrations from50ug/ml and300ug/ml.2.If the concentration of BCG is10ug/ml,it shows a growing tendency oftyrosinase activity,but thymosin makes the tyrosinase more active at300ug/ml,andthymosin have a better effect on tyrosinase than BCG.3.As concentration of BCG changing slightly increases,it shows a growing tendencyof tyrosinase activity,the thymosin promotes the effect of cell melanin content,andthere are time and dose-dependent relationship,in which processing to300ug/ml72hours of the most obvious role of melanin content increased.Conclusion:A certain concentration of both BCG and thymosin A375can promotemelanocyte proliferation,increased cell tyrosinase activity, which lead to achieve theeffect of promoting melanin synthesis, and their catalytic effect with the treatmenttime and dose-associated.
