Experimental Studying The Effect Of Chinese Herbal Medicine KBLS On Melanocyte Metabolism

Objective:By studying the effects of Kang Bai Ling(KBLS) on proliferation, activity of tyrosinase and melonogenesis in B16murine melanoma cells in vitro to explain the relationship between melanocyte metabolism and KBLS, describing these mechanisms in the treatment of vitiligo.Methods:B16murine melanoma cells were cultivated in vitro with five concentrations medicines. The blank group only had DEME and the control group only had melanoma cells. The cytological and enzymological methods were be used for studying the effects of KBLS on proliferation, activity of tyrosinase and melonogenesis in B16murine melanoma cells. The MTT was used to detect the proliferation of melanoma cells, enzymological method was used to measure the activity of tyrosinase and melanin could be directly detected by its absorption at405nm.Results:1. The water extract of KBLS can proliferate B16murine melanoma cells obviously. Five test groups were significantly different from the control group(P<0.05).ALL of them showed the positive corrlation. The higher concentration (15～25mg/ml) showed advanced action, while the lower concentration (5～10mg/ml) showed slight inhibition.2. The water extract of KBLS can enhance activity of tyrosinase. The results of KBLS on the tyrosinase activity showed that test groups which were different from the control group (P﹤0.05) could significantly enhance the activity of tyrosinase. All the groups showed the positive correlation. The murine melanoma cells cultivated in vitro with KBLS for24hours had optimal activation effects, especially the lower concentration(5～15mg) had a better effect than48hours or72hours(P<0.05).3. The water extract of KBLS can accelerate melanogenesis. Compared with the control group(P﹤0.05),five test groups can enhance melanogenesis in the B16murine melanoma cells. But the effect showed the negative correlation in statistics, although the higher concentration was just a bit worse than the lower concentration.4. The alcohol extract of KBLS can proliferate B16murine melanoma cells obviously. Five test groups were significantly different from the control group(P<0.05).All the groups showed the positive correlation.5. The alcohol extract of KBLS can enhance activity of tyrosinase. The results of KBLS on the tyrosinase activity showed that test groups which were different from the control group (P﹤0.05) could significantly enhance the activity of tyrosinase. All the groups showed the positive correlation. The murine melanoma cells cultivated in vitro with KBLS for48hours had a better effect than48hours or72hours(P<0.05).6. The alcohol extract of KBLS can accelerate melanogenesis. Compared with the control group(P﹤0.05),five test groups can enhance melanogenesis in the B16murine melanoma cells with a positive correlation.Conclusion:Chinese medicine KBLS can proliferate B16murine melanoma cells, enhance the activity of tyrosinase and accelerate melanogensis.As the increasing of concentration, the effects were enhanced roughly.
