Objective:By detecting the expression differences of Ackl gene in gastric cancer tissues and para-carcinoma tissue,normal gastric tissue, analyze the relationship between Ackl and gastric cancer patients’gender, age, tumor location, tumor differentiation, TNM stage and provide the basis for diagnosis and treatment of gastric cancer.Methods:Collected surgical resection and confirmed by pathological paraffin specimens,gastric cancer60cases as gastric cancer group,collect tissues adjacent to cancer15cases and normal gastric tissuse15cases as adjacent gastric group and normal group.Immunohistochemistry streptavidin peroxidase(SP) was used to detect protein of Ackl in gastric group,adjacent gastric group and normal group.Respectively collected14cases of patients with cancer tissue, the tissue adjacent to cancer and normal tissues, and divided into14groups.Reverse transcription polymerase chain reaction(RT-PCR) was used to detect mRNA of Ackl in14cases fresh specimens gastric group,adjacent gastric group and normal group.Results:(1)Ackl protein the positive rate expression in gastric cancer tissues is high(87%),part of the expression in the adjacent to carcer tissuse,no or lower expression in normal tissuse,this difference is significant(P=0.003<0.05).(2)Comparing two groups think that normal stomach tissue and gastric cancer tissue,stomach tissue and cancer tissue adjacent to Ackl protein expression are statistically significant differences (P=0.000、 P=0.002<0.05),but the normal stomach tissue and cancer of the stomach cancer tissue adjacent to Ackl protein expression differences were not significant (P=0.054>0.05)(3)The positive rate Ackl mRNA in gastric cancer [1.1475(1.0940,1.4380)] is higher in adjacent gastric cancer tissue [2.4538 (2.1294,2.8795)]and normal tissue[13.1326(11.8353,15.2316)],and this difference is significant.(4)Ackl expression and gastric cancer patients about the differentiation degree (P=0.017)and TNM staging(P=0.022),the low differentiated the lack of the higher rate of expression;Ackl protein expression and patient age(P=0.168),sex(P=0.170) and tumor location(P=0.549) have nothing to do.Conclusion:(1)In the gastric cancer tissues,the Ackl protein and Ackl mRNA expression positive rate are high,is there a difference with adjacent normal tissues and normal tissues.(2)Ackl protein expression positive rate is associated with tumor differentiation and TNM stage, its in the process of gastric cancer occurrence and development plays an important role.(3)Ackl protein expression were positively correlated with tumor differentiation and TNM stage, the lower degree of differentiation and the higher TNM stage, the higher positive rate of Ackl protein expression.
