Effect Of The Qianyang Jiedutongluo Drink Containing Serum AngⅡ Induced TLR4Signaling Pathway Of Neonatal Rat Cardiac Myocyte Hypertrophy

Purpose: Through using the method of Cell cuture, to research of QianyangJieDuTongLuo drink mixture (XFJD) inhibition of cardiac myocyte hypertrophy andto explore its mechanism.Method: The research adopts the method of the serum pharmacology, in vitrocultures rat myocardial cells, divided into normal control group, model group,the Qianyang JieDuTongLuo party group, valsartan group, pioglitazone group. Andmethyl thiazolyl tetrazolium (MTT) assay to detect myocardial cell’sproliferation inhibition. The reaction assay with reverse transcriptionpolymerase chain detects c-fos and c-myc mRNA expression. Transmission electronmicroscopy observes cell ultrastructure. Immunohistochemical staining to detectthe expression of TNF-a, TRL4.Result: AngⅡ roles in cells after12hours, compared with the normal controlgroup, c-fos and c-myc mRNA expression was significantly increased (P <0.05),myocardial cell proliferation (P <0.05). Compared with the model group, QianyangJieDu TongLuo drink after the intervention, the myocardial cells proliferationwas inhibited (P <0.05), c-fos, c-myc mRNA downregulation (P <0.05).The resultsshow that electron microscopy myocardial ultrastructure is better with the effectof the Xifeng detoxification tongluo endocytosis, with best Chinese medicine10%of the dose effect. Expression of Qianyang Jiedu Tongluo Decoction inhibitmyocardial cell TLR4, TNF-a factor.Conclusion:Qianyang detoxification Tongluo drink cansuppressAngⅡ-inducedcardiac myocyte proliferation. It has the function of Anti-cardiomyocytehypertrophy.
