Effect Of Ziyin Qianyang Recipe On Myocardial Remodeling In 2K1C Hypertensive Rats Based On Rho/Rock Pathway

Objective:The relationship between Rho/Rock pathway,TGF-?2 and Ang?and cardiomyocyte remodeling was explored.And the effects of Ziyin Qianyang Recipe on Rho/Rock signaling pathway in myocardial cells of 2K1 C rats were observed.This experiment enriches the mechanism theory of Ziyin Qianyang Prescription in treating diseases.Methods:Sixty 8-week-old healthy homologous male SD rats(180±20g)of clean grade were selected and fed adaptively for one week.Then 48 rats were randomly selected to establish two kidneys and one clip renal hypertension model.The remaining 12 rats were sham-operated group.Thirty-six rats were randomly divided into model group,traditional Chinese medicine group and western medicine group.Sham operation group and model group were given saline infusion;The traditional Chinese medicine group was given Ziyin Qianyang Decoction(30g/kg/d)and the western medicine group was given enalapril(1mg/kg/d)according to the standard ratio and then gavaged for 8 weeks.The systolic blood pressure of caudal artery was measured before,2 weeks after operation,2 weeks after administration,4 weeks after administration and 8 weeks after administration.After 8 weeks of administration,rats were sacrificed and abdominal aorta blood was taken to detect the expression of Ang?.The left ventricular wall myocardium was taken and the content of TGF-?2 in myocardium was detected by immunohistochemical method.The content of Rho A-m RNA and Rock1-m RNA were tested by real-time fluorescence quantitative PCR.Rho A and Rock1 were determined by Western Blot method.Histopathological changes of myocardium observed by HE staining.Under the condition of Mason staining,we observed the changes of collagen tissue.Results:After 2 weeks of modeling,the blood pressure in model group,traditional Chinese medicine group and western medicine group increased significantly compared with sham operation group(P<0.05).After 8 weeks,compared with the model group,the blood pressure values of both the traditional Chinese medicine group and western medicine group decreased(P <0.05).Compared with sham operation,LHWW/BW in model group increased significantly(P <0.01).Compared with the model group,the LHWW/BW of two treatment groups decreased significantly(P <0.01).Compared with sham operation group,the concentration of Ang? in the model group and two treatment groups(P <0.05).Compared with the model group,the expression of Ang?in two treatment groups decreased significantly(P <0.01).Compared with sham-operated group,the expression of TGF-?2 in model group increased significantly(P <0.01).Moreover,myocardial tissue slices showed that the positive expression in model group was significantly higher than that in sham operation group,while the positive expression in two treatment groups was significantly lower than that in model group.Compared with the sham-operated group,the levels of Rho A-m RNA,Rock1-m RNA and the contents of Rho A and Rock1 protein in the model group were significantly increased(P <0.01).Compared with sham-operated group,the levels of Rho A-m RNA,Rock1-m RNA and the contents of Rho A and Rock1 protein in model group increased significantly(P <0.01).Compared with the model group,the levels of Rho A-m RNA,Rock1-m RNA and the contents of Rho A and Rock1 protein in the traditional Chinese medicine group and the western medicine group were significantly lower(P <0.01).In HE stained sham operation group,the intercellular space of myocardial cells was more obvious,the cytoplasm was clearly stained,and the fibrous tissue was not significantly increased.In the model group,myocardial cell gap disappeared,cytoplasm staining was not clear,fibrous tissue proliferated,some cells arranged irregularly,myocardial tissue proliferated and other myocardial remodeling phenomena.And in the Masson staining model group,the nucleus and cytoplasm of cardiac myocytes were not clearly stained,the blue collagen tissue was abundantly expressed,and some cells were irregularly arranged.However,the phenomenon of myocardial remodeling in the two treatment groups was significantly reduced compared with the model group.Conclusion:Ziyin Qianyang Decoction can effectively and gently reduce blood pressure and the expression of Ang ?,TGF-?2 and collagen,and improve the phenomenon of myocardial remodeling in rats.Ziyin Qianyang Decoction can effectively inhibit the activation of Rho A/Rock1 signaling pathway.
