Objective:To investigate expression characteristics and variation of DEC1and TIAM1in oral squamous cell carcinoma (OSCC), to explore the correlation between expression of DEC1, TIAM1and recurrence of OSCC at the level of gene and protein; providing the reference of assessment of the prognosis of OSCC.Method:1. Expressions of DEC1and TIAM1in OSCCWe collected80cases, their diagnosis were OSCC (including high, middle and lower differentiation groups, grade Ⅰ-Ⅳ, and they didn’t receive radiotherapy and chemotherapy preoperation). All cases collected from department of maxillofacial surgery, Xiangya Hospital of Central South University during from2010to2012, and20cases of normal oral mucosa. Immunohistochemical method was applied to detect the expression of DEC1protein and TIAM1protein.2. Correlation between expressions of DEC1, TIAM1and the OSCC recurrence and metastasis.We collected56cases of the tumor, their diagnosis were OSCC (including high differentiation group, grade III-IV, and they didn’t receive radiotherapy and chemotherapy preoperation), in the department of maxillofacial surgery, Xiangya Hospital of Central South University during from2009to2012. The expression of DEC1protein and TIAM1 protein in56specimens were detected by Immunohistochemical method. The Image-Pro Plus6.0image analysis technique was applied to detect the number of the positive units (IOD value) from the expression of DECI protein and TIAM1protein.We collected32cases of the tumor, which were frozen by using liquid nitrogen and were diagnosised as OSCC (including high differentiation group, grade Ⅲ-Ⅳ, and they didn’t receive radiotherapy and chemotherapy preoperation), in the department of maxillofacial surgery, Xiangya Hospital of Central South University during from2009to2012. The expression of DEC1mRNA and TIAM1mRNA were detected by RealTime PCR.All data was analysised by using SPSS19.0statistical software.Result:1. Expressions of DEC1and TIAM1in OSCC.The result of immunohistochemistry detection showed that positive expression rates of DEC1and TIAM1in OSCC were92.50%and91.25%respectively. In normal oral mucosa tissues, positive expression rates of them were5.00%and10.00%respectively, The difference was statistically significant (P<0.05). In OSCC, the difference in expression of DEC1and TIAM1were statistically significant (P<0.05) between high differentiation group and low differentiation group, between lymph node metastasis group and non-lymph node metastasis group.2. Correlation between expressions of DEC1, TIAM1and the OSCC recurrence and metastasis.The result of immunohistochemistry detection showed that the difference in expression of DEC1and TIAM1was statistically significant (P<0.05) among different OSCC groups (high differentiation group, grade III-IV). The expression of DEC1, TIAM1in1-year recurrence group was significantly higher than other groups. The expression of DEC1and TIAM1in3-years no recurrence group was the lowest.The result of RealTime PCR detection showed that the content of DEC1mRNA and TIAM1mRNA in OSCC cells were higher than that in normal group. The comparison study among different OSCC groups has statistical significance (P<0.05). The expression of DEC1mRNA and TIAM1mRNA in1-year recurrence group was significantly higher than the other groups. The expression of DEC1mRNA and TIAM1mRNA in3-years no recurrence group was the lowest.Conclusion:1. DEC1and TIAM1were located in cytoplasm of oral squamous cell carcinoma cells, showing higher expression than in normal oral mucosa.2. The expression of DEC1mRNA, TIAM1mRNA in1-year recurrence group of OSCC were significantly higher than the other groups. The expressions of DEC1and TIAMl were negatively correlated to the prognosis of OSCC.
