Objcetive:(1) To verify the previous chip results of our research group by studying the expression of MAL and PAI-1Gene and explore the relation between the genes and the oral verrucous carcinoma’s development from Gene level and protein level;(2) Try to elucidate the pathogenesis of oral verrucous carcinoma from the corner of molecular genetics and support the evidence for the diagnosis and treatment of oral verrucous carcinoma. Methods:To use RealTime-PCR method detected five cases of oral verrucous carcinoma cancer and adjacent normal tissue in five cases of oral squamous cell cancer and adjacent normal tissue of the MAL and PAI-1of gene expression levels, to verify the results of the early chip;(2) To use of immune histochemical method detected MAL and PAI-1of oral verrucous carcinoma (17cases), oral squamous cell carcinoma (17cases), normal oral mucosa (17cases) in the distribution, and statistical analysis;(3) To use of Western Blot detected three cases of oral verrucous carcinoma, three cases of oral squamous cell carcinoma three cases of normal oral mucosa in MAL and PAI-1of gene and protein expression to the relationship between discussion and oral verrucous carcinoma. Results:MAL expression in oral verrucous carcinoma was significantly lower than the normal control group, the expression of PAI-1in oral significantly higher than the normal control group, consistent with previous gene chips; results suggest MAL and PAI-1of genes and oral verrucous of cancer development;(2) MAL protein low in oral expression, oral verrucous carcinoma tissue expression of PAI-1verify MAL and PAI-1can be used as the reference diagnostic indicator of oral verrucous carcinoma; gene transcription and translation level of consistency. Conclusion:MAL gene expression in oral verrucous carcinoma was significantly lower than the normal control group, the results are consistent with the previous gene chips of our group, but MAL gene and oral squamous cell carcinoma also need to explore further; PAI-1gene in oral squamous cell carcinoma than in the corresponding normal tissues were significantly increased in oral expression relative to their normal control group, although an increasing tendency, but not statistically significant, need further verification;(2) MALprotein expression in oral verrucous carcinoma and oral squamous cell carcinoma compared with normal mucosa showed low expression, but no significant difference in oral verrucous carcinoma and oral squamous cell carcinoma; PAI-1protein in oral cancer and oralexpression in squamous cell carcinoma than normal mucosa showed high expression, but no significant difference in oral verrucous carcinoma and oral squamous cell carcinoma.
