Objective:Hydroxyapatite nanoparticles (nHAP) were prepared by a chemical coprecipitation-hydrothermal synthesis. Study on the effects of growth of nanoparticle with various factors. To improve the transfection efficiency of vitro cells by modified nHAP and surface modification with Polyethylenimine(PEI).Methods:By means of transmission electron microscopy observation and Size、Zeta detection after surface modification with PEI, the modified nHAP/PEI-nHAP were determined for their capacity of suspension stability, DNA binding and protection, transfection efficiency of vitro cell.Results:1)The experiment result indicate that the optimal Ca/P ratio was1.67:1, both the concentrations of Ca(NO3)2·4H2O and (NH4)2HPO4were0.15mmol/L, the rate of mixing was15drop/min, the pH value was between10and10.5, the temperature was170℃, reaction for3hours, the aging time was24hours.2) the modified nHAP after surface modification with PEI had better suspension stability, DNA binding and transfection efficiency.Conclusion:1)The morphology structure and thermal stability of nanoparticle were affected by Ca/P ratio, concentration, rate of mixing, pH value, temperature, reaction time,and aging time.2)when the PEI concentration was4‰, PEI-nHAP had better. DNA binding stability.3) The transfection efficiency of the modified nHAP was approximately6%, and the transfection efficiency of the modified nHAP by PEI was approximately21%.(25figures,34tables,31references)...
