Objective:To investigate the efficiency of hydroxyapatite nanoparticles by polyethylenimine(PEI) (PEI-nHA) mediationg p53 into HNE2 nasopharyngeal carcinoma cells.Methods:(1) Construction of pEGFP-C1-P53 vector. (2) Hydroxyapatite nanoparticles by polyethylenimine(PEI) (PEI-nHA) and LP2000 were used to mediate p53 transfecting into HNE2 nasopharyngeal carcinoma cells, the efficiency of transfection is detected.(3) RT-PCR and Western-blot were used to investigate the expression level of p53.Results:Green Fluorescence was not detected in blank group, while in LP2000 group, about 40% cells were detected, in PEI-nHA group, when adding 62.5ug/ml PEI-nHA,the green fluorescence were detected only in 4% cells, while adding 125ug/ml PEI-nHA,the rate was 6%, and when the concentration increase to 250ug/ml, the rate was 5%, and the concentration increase to 500 ug/ml, the rate was 1%. RT-PCR and Western-blot experimentation show the expression of P53,but the expression of P53 in LP2000 group was higher than that of PEI-nHA group.Conclusion:In this experimentation, Compared with Lp2000, PEI-nHA is less efficient in mediating transfection of P53 into HNE2 cells.
