| Objective:According to rhinitis:spray the attending functions, through the establishment of the experimental animal model, further explore the mechanism of the treatment of allergic rhinitis. SP (substance P), TNF alpha (tumor necrosis factor alpha), Annexinl associated protein1(membrane) and VCIM-1(blood cell adhesion molecule-1) for the experiment research direction, content and target cells under light microscopy observation number difference, which validate the mechanism of action of treatment of allergic rhinitis, and to provide basis for clinical rational drug use.Methods:70healthy guinea pigs were selected, randomly divided into seven groups, with10rats in each group, respectively, the blank control group, model group, rhinitis Tongqiao spray high, medium and low dose group, the Ditong rhinitis water group and triamcinolone acetonide spray group. All experimental groups except blank control group, with the10%TDI olive oil solution as the molding material, the nasal drops drip into the sensitizing agent, so that the formation of nasal septum mucosa inflammation, establish allergic rhinitis animal model, to observe and record the change of each group of guinea pigs were determined in each group; SP, the nasal mucosa of guinea pigs TNF-α, Annexinl, VCIM-1by the method of content determination of cell number; IgE and IL-4each guinea pig serum by ELISA method; the pathological changes of nasal mucosa HE staining method.Results:Rhinitis Tongqiao spray can improve the guinea pig allergic rhinitis symptoms of inflammation. Behavioral scores, compared with the model group, score decreased significantly, the difference was statistically significant (P<0.05or P<0.01); content of Tongqiao rhinitis spray group and the control group of Western medicine in nasal mucosa in SP was significantly lower than the model group, the difference was statistically significant (P<0.01), compared with the Western Medicine control group, no significant difference the content (P>0.05); content of Tongqiao rhinitis spray group and Western medicine control group, TNF-αin nasal mucosa was significantly lower than that of model group, the difference was statistically significant (P<0.01or P<0.05), compared with the control group, the content had no significant difference (P>0.05); the content of Annexinl in the nasal mucosa of control group rhinitis Tongqiao spray group and Western medicine was higher than that of the model group, the difference was statistically significant (P<0.05or P<0.01), compared with the control group, no significant difference (P>0.05); the content of rhinitis Tongqiao sprays and control the content of VCIM-1in the nasal mucosa was significantly lower than that of model group, the difference was statistically significant (P<0.01or P<0.05); compared with western medicine the control group had no obvious difference, the content (P>0.05). The model group was significantly higher than that of serum IgE concentration of rhinitis Tongqiao sprays in each dosage group and the other two positive control group (P<0.01or P<0.05); the model group was significantly higher than that of serum IL-4concentration of rhinitis Tongqiao sprays in each dosage group and the other two positive control group (P<0.01or P<0.05)Conclusion:Tongqiao rhinitis spray to treat allergic rhinitis, reduce the content of IgE and IL-4in serum, decrease the expression of SP, TNF-α, VCIM-1in nasal mucosa, and up-regulate the expression of Annexinl in nasal mucosa and nasal mucosa, reduce congestion, edema, inflammatory symptoms, therapeutic effect of triamcinolone acetonide spray compared with no obvious difference. This shows that higher levels of rhinitis Tongqiao spray in regulation of inflammatory mediators display. |
