Objective:To study the chemical constituents and biological activity in the ethyl acetateextraction of Dendrobium thyrsiflorum;To investigate polysaccharide and mannosecontent and the characteristic spectrum in different origin of Dendrobium officinale andinitial quality of it was evaluated.Methods:Using silica atmospheric column chromatography, reverse phase columnchromatography(RP-18),hydroxyproyl glucose gel(Sephadex-LH-20) columnchromatography and other chromatographic separation material,Using massspectrometry,nuclear magnetic resonance spectroscopy determine the structure ofmonomer compounds in D. thyrsiflorum;Using MTS filter antitumtor activity ofcompounds1,2,3and4;Using UV and HPLC determine polysaccharide and mannosecontent and the characteristic spectrum in different origin of D. officinale.Results: Isolated and identified a total of16compounds in D. thyrsiflorum,compound3has significant antitumtor activity;Using Phenol-Sulphate Acid method to determine thecontent of polysaccharide in different origin of D. officinale.Polysaccharide content ashigh as50.07%in chang ning,the lowest is23.49%in tian tai,both differ by17.16%.Theaverageof polysaccharide content is40.79%in14origins of yunnan,the average is28.46%in foreign languages;Establish the HPLC method and determine mannosecontent in different origin of D.officinale.Mannose content as high as36.41%in jinggu,the lowest is20.64%in tian tai,both differ by20.64%.The averageof polysaccharidecontent is28.97%in14origins of yunnan,the average is22.60%in foreign languages;The chemical constituents of D.officinale were optimally separated,among which26fingerprint peaks in common were confirmed.The overall view of the peak appearancefingerprinting basically the same.In addition to jing gu and zhenyuan is0.835and0.826,the remaining similarity origin were above0.9from similarity evaluation results.Conclusion: Compund1、3、4、8~16were isolated from D. thyrsiflorum for the firsttime;Suger content were vary widely in different origin of D. officinale,the reason may be related to planting environment、growth conditions、growth years and plantgenetic characteristics,which needs futher study.The chemical composition is nodifference in kind,but in peak area chemical composition have some difference incontent of the same harvesttime in different origin of D. officinale.
