Objective:Research on the excavation of different chemical compositions of Wild-simulated D.officinale by UPLC Q-TOF/MS and the difference of polysaccharide content of D.officinale from various areas has been compared.Methods:1.Collection of D.officinale samples: In accordance with factors such as collecting place,altitude and phototropism.2.Determination of polysaccharide content of D.officinale: According to Chinese Pharmacopoeia(2015),polysaccharides were extracted by water extraction and alcohol precipitation,and the polysaccharide content was measured by phenol sulfuric acid color reaction.3.The analysis of secondary metabolites of D.officinale: UPLC Q-TOF/MS was used to study the secondary metabolite profiles of D.officinale.4.The correlation between components and targets were supported by Network pharmacology.Potential targets for the different chemical constituents in D.officinale,PPI network analysis,KEGG pathway and disease analysis and other network pharmacological methods were performed to predict potential targets and corroborate the correlation between the chemical constituents and pharmacological effects.Results:1.According to Chinese Pharmacopoeia(2015),the polysaccharide content of D.officinale was detected.The elevation factor had the most significant effect on the polysaccharide content of D.officinale,which was higher at very low elevations.In areas close to the high-altitude areas where D.officinale was reproduced in Flora of China,the polysaccharide content of D.officinale collected at an altitude of 918 meters was the highest.Light factors also play a role,but to a lesser extent.2.The planting altitude of D.officinale in Xingyi,Guizhou was the closest to the propagation altitude of D.officinale described in Flora of China,but the content of polysaccharide was the lowest.In order to find out whether there were relevant superior chemical components in the wild D.officinale under the propagation conditions.D.officinale in Xingyi,Guizhou was used as the model sample,and OPLS-DA method was used for comparison and analysis with samples from Dushan,Guizhou,Danzhai,Taizhou,Zhejiang,and Yueqing,Zhejiang.The results showed that 12 small molecule components were dominantly expressed in Xingyi samples at the breeding elevation of the near-wild D.officinale.The structure was inferred by control chromatography and high-resolution molecular weight comparison,secondary fragment ion comparison,literature and database comparison,etc.3.To investigate whether the above 12 chemical components were related to pharmacological and pharmacological effects,using its three-dimensional chemical structure,the targets were predicted by Swiss Target Prediction database,and related diseases were predicted based on the target function.The results show that 11 of the 12 different chemical components have a total of 354 potential targets.These acting targets were involved in related diseases in which neurological disorders predominated.Conclusion:The main factor affecting the polysaccharide content of D.officinale was altitude.The high-resolution mass spectrometry revealed that the wild D.officinale under nearaltitude conditions has 12 different chemical components,and its abundance was in the opposite trend to the polysaccharide content of D.officinale.This provides data to support the development of pharmacologically oriented quality standards for D.officinale.
