| In this research,the raspberry leaves was taken as the research material.the extracting technology of ethanol extract in Rubus idaeus Leaves was optimized. The ethanol extract was separated into three subfractions by organic solvents of different polarities based on liquids-liquids extracting method. The different antioxidant systems were chosen to evaluate comprehensively the antioxidant activities of ethanol extract and the fraction of different polarity. In vivo antioxidant effects were studied by choosing D-gal aging mice.A preliminary study of its chemical composition was made.Experiments were carried out by thermal extraction of single factor and orthogonal experiments to determine the raspberry leaf flavonoid extract the optimum conditions:70%ethanol, extraction temperature60℃, extraction time of1h, solid-liquid ratio1:25, three times under the optimum extraction, raspberry leaf flavonoids in the crude yield31.15%. And crude extract of raspberry leaf method and further purified by solvent extraction, different solvents were used to extract extraction, petroleum ether fractions obtained, ethyl acetate fraction, n-butanol fraction and water-soluble components. By flavonoids, after extraction of the ethyl acetate fraction flavonoid content, it is up to300.82mg rutin/g.and significantly higher than the other phase.In this paper, a series of in vitro oxidation experiments, comparing the various components of raspberry leaf extract in vitro antioxidant activity.The results showed that different polarity components have shown antioxidant and free radical scavenging activity. Determination of total reducing power, FRAP method antioxidant capacity, ability to remove ABTS radicals, DPPH radicals, and the ability of superoxide anion, Fe ion chelating ability, inhibition of mouse liver homogenate lipid peroxidation:ethyl acetate phase> n-butanol phase> crude extract> aqueous> petroleum ether phase.Ethyl acetate fraction and the crude extract can inhibit the hemolysis of red blood cells, the effect of ethyl acetate fraction was significantly higher than the crude extract. Different parts of the polar extract flavonoids and antioxidant capacity were positively correlated.At the end of experiment of feeding mice, the behavioral tests including the water maze test in mice, tight rope experiments and field experiments showed that mice administered cognitive learning, space exploration capabilities.as well as memory and physical are significantly stronger than D-galactose induced aging mice mice.Selection of D-galactose induced aging mice to study the raspberry leaf ethyl acetate extract in mice in vivo antioxidant systems. The results showed that:0.09%dose group (dose group II),0.18%dose group (dose group Ⅲ).0.25%dose group (dose group Ⅳ) can significantly or very significantly increased in mice serum, liver and brain tissue SOD. T-AOC and GSH-px vitality, reduce the MDA content (P<0.05or P<0.01). Little effect on CAT activity, between dose group and model group is not very significant difference.Mouse liver tissue SODmRNA quantitative PCR results show that:treatment group SOD1gene expression in mouse liver tissue was significantly higher than model group (P <0.05or P<0.01).In this paper ethyl acetate extract of raspberry leaf antioxidant components of functional components of a preliminary analysis by HPLC analysis of a variety of conditions, the ethyl acetate extract of raspberry leaves a group of isolated components of peak.Rutin as standard, may be inferred that the main functional components of rutin However, a wide range of flavonoids contained in complex and require further isolate and study. |
