Objective: To investigate the efficacy of Tang Ji Kang ointment and the gel ofrhEGF (Recombinant Human Epidernal Growth Factor) and expression ofCTGF (Connective Tissue Growth Factor)、 IGF-I (Insulin-like GrowthFactor-I)、PDGF-AA (Plateled Derived Growth Factor) in rat diabetic skinulcers for one or two weeks,which will provide the experimental basis formechanism of Tang Ji Kang ointment and the gel of mrhEGF in the treatment ofdiabetic skin ulcer and clinical application. Methods:1. Modeling grouping:60model diabetic skin ulcer rats were randomly divided into5groups: blankcontrol group,model control group, Tang Ji Kang control group, growth factortreatment group, self control group. From the following day of the modelingsuccess, every group will be dressing until the wound healing and observe thetreatment effect.2. Draw materials and staining: after one week and two weeksdressing the wound was drawn, making routine paraffin section, then displaythe pathological changes and immunohistochemical expression of CTGF, IGF-Ⅰ, PDGF-AA using HE staining.3with IPP (Image-Pro Plus)6software forimage analysis of statistical data: the data of4standard deviations (x±s) said,the data were analyzed by SPSS13statistical software, the differences betweengroups were analyzed by t-test, when P <0.05there was a significant differencebetween the standard.Results:①the animal model group: size, color, stem growth of humidity and granulation tissue, results show that Tang Ji Kanggroup heal faster, the velocity of healing is growth factor group, the blankgroup compared with model group, the healing speed slightly faster.②Observed HE staining, the connective tissue, fibroblasts and granulation tissuein Tang Ji Kang group grow faster than the growth factor group. I n growthfactor group they grow faster than in model group and blank group.thehealing speed of blank group and model group is the worst; Growth factorgroup arranged slightly worse than Tang Ji Kang whose fibroblasts arrangedmore orderly.③The average optical density by immunohistochemicaldetection of groups one and two weeks CTGF, IGF-Ⅰ and PDGF-AA value,results showed that the corresponding period CTGF in Tang Ji Kang group andwhich in other groups had significant difference (P<0.05).Tang Ji Kang groupaverage optical density value was minimum.Growth factor group was slightlyhigher than in the Tang Ji Kang group,while the blank group was higher thanthat of treatment two groups.And model group was the highest. IGF-Ⅰof TangJi Kang group and the other groups had significant difference (P<0.05) overthe same period, when the Tang Ji Kang group average optical density value ofminimum, growth factor group is slightly higher than it in Tang Ji Kang group,the blank group was higher than that of treatment two group, model group wasthe highest. Tang Ji Kang group and the other groups’s PDGF-AA weresignificant difference (P<0.05) over the same period, when the Tang Ji Kanggroup average optical density value of minimum, growth factor group was slightly higher than in Tang Ji Kang group.and the blank group was higher thanthat of treatment two group,while model group was the highest.④The averageoptical density of self control group revealed: Tang Ji Kang treatment groupwas lower than in the right growth factor treatment group and there weresignificant differences (P<0.05). Conclusion:①Tang Ji Kang group treatmentresult who had less scar formation is faster than growth factor group treatmentresult, which may be related to the effect of expression of skin CTGF, IGF-Ⅰand PDGF-AA.②Tang Ji Kang whichis a new drug for treatment of diabeticskin ulcer, the clinical curative effect is better than rhEGF (recombinant humanepidermal growth factor)。...
