objective:Knee cartilage injury is more common in clinical practice, but its ability to self-repair is extremely limited. It lack of a precise and effective treatment and likely to develop traumatic arthritis and cause joint dysfunction, and so has been the difficulty of the medical field and research focus. This subject apply Shao-yang Prescription intervene with cartilage damage in vivo in rats and observe the repair of articular cartilage injury and confirmed its repairment of cartilage defects.It can explore its part mechanism of the treatment of cartilage damage and provide strong experimental evidence for the application of Chinese medicine theory,open new avenues for the treatment of cartilage damage. Lay the foundation for the next step to continue to study the mechanism of Shao-yang Prescription repair cartilage damage and elucidate the role of the Prescription from the the molecular Biology and Cytology level, provide strong evidence for clinical promotion. Methods:1.Knee cartilage damage in animal model and drug intervention:Select6to8weeks clean SD rats fed under the same conditions (Luzhou Medical Experimental Animal Center Provided)27, male or female, weight250±20g. Weighed and randomly assigned into three groups, according to the group balanced consistent principle,scilicet Shao-yang Prescription the experimental group (S group), Glucosamine Hydrochloride capsule group (A group) control group,blank group (K group), n=9. In sterile conditions,drill a hole in the femoral trochlear middle with a hollow needle whose diameter is2mm and then form animal models of a full-thickness cartilage damage. The day after the completion of the modeling, began to gavage drugs:the S group Shao-yang Prescription gavage,A group Glucosamine Hydrochloride capsule gavaged,normal saline group K.2.To evaluate curative effect of Glucosamine Hydrochloride capsule and Shao-yang Prescription which affect the level of IL-1β in joint fluid on rats after articular cartilage injury in different periods (4weeks,8weeks,12weeks).3.To evaluate curative effect of Glucosamine Hydrochloride capsule and Shao-yang Prescription which affect cartilage tissue morphology and matrix Ⅰ、Ⅱ phenotypic on rats after articular cartilage injury in different periods (4weeks,8weeks,12weeks).Results:(1)Three sets of synovial fluid in comparison with the level of IL-1βdetection, S and A:8and12weeks of group Ais higher than the S group (P<0.05);8and12weeks K group high8and12weeks K group was higher than in group S (P<0.05); A group (P<0.05). Own comparison before and after treatment:S group first eight weeks,12weeks is less than4weeks (P<0.05); The A group8weeks,12weeks were lower than the first4weeks (P<0.05); K group4weeks8weeks12weeks (P<0.05).(2)General observation, histological analysis and transmission electron microscope examination showed the S group A cartilage defect repair tissue quantity and quality is better than the K group and S group effect is stronger than Group A repair.(3)Shao-yang Prescription and Glucosamine Hydrochloride capsule can improve the expression of type Ⅱ collagen in cartilage tissue repair after injury (P<0.05), but the two groups is not significant (P>0.05).
