Objective:To observe the damage on Human Umbilical Vein Endothelial Cells(HUVEC) in vitro induced by intermittent triglyceride and High Glucose joint training and discuss the damage to vascular endothelial of postprandial hypertriglyceride, intermittent triglyceride, coexistence of hyperglycemia and hypertriglyceride.Methods:1 HUVEC were used at the same passage(sixth) and left to grow in complete medium without any serum for 24 h for treatments.2 Collect Normal serum and high triglyceride sera from hospital, mix them to corresponding concentrition and then inactivate and sterilize them. The other target in serum such as TC, GLU, LDL, HDL were mixed to similar concentration, and measured in hospital.3 The experiment was divided into 10 groups, Gave the following conditions for 3 days: NO.1 group add 5.5 mmol/L glucose and 1.56 mmol/L triglyceride serum, NO.2 group add 5.5 mmol/L glucose and 3.45 mmol/L triglyceride serum, NO.3 group add 5.5 mmol/L glucose and 5.52 mmol/L triglyceride serum, NO.4 group add 5.5 mmol/L glucose and 1.56 mmol/L(8h)/3.45 mmol/L(16h) triglyceride serum, NO.5 group add 5.5 mmol/L glucose and 1.56 mmol/L(8h)/5.52 mmol/L(16h) triglyceride serum, NO.6 group add 25 mmol/L glucose and 1.56 mmol/L triglyceride serum, NO.7 group add 25 mmol/L glucose and 3.45 mmol/L triglyceride serum, NO.8 group add 25 mmol/L glucose and 5.52 mmol/L triglyceride serum, NO.4 group add 5.5 mmol/L glucose and 1.56 mmol/L(8h)/3.45 mmol/L(16h) triglyceride serum, NO.5 group add 5.5 mmol/L glucose and 1.56 mmol/L(8h)/5.52 mmol/L(16h) triglyceride serum.And then gave each group add 5.5 mmol/L glucose and 1.56 mmol/L triglyceride serum for 8h. Measurate the contents of monocyte chemoattractant protein-1(MCP-1), vascular cell adhesion molecule(VCAM),interscular cell adhesion molecule(ICAM), C-reactive protein(CRP), malondiadehyde(MDA), superoxide sismutase(SOD), nitric oxide(NO) and MTT. MTT for each group was measured.4 All the data were dealed with statistical software packages SPSS13.0, the data were expressed as the mean±standard deviation( x ±s), the data were analyzed using Oneway ANOVA of completely rabdom design, multiple comparisons were analyzed using LSD test. Statistical significance was set at P<0.05.Results:1 MTT:The OD of MTT in the group with middle concentration of triglyceride fluctuation was higher than the group with high concentration of triglyceride fluctuationod. With the same concentration of glucose, compared with the low concentration of triglyceride, the OD of the groups in middle triglyceride concentration of triglyceride groups were higher, while the OD of high concentration groups were lower. With the same concentration of triglyceride, the OD of the groups with high concentration of glucose were higher than the groups with low concentration of triglyceride. The OD of onefold high concentration glucose groups were higher than the groups with onefold high triglyceride, but were lower than the groups with onefold middle concentration of triglyceride. While the OD of the group with high concentration of triglyceride and high concentration of glucose joint was the lowest. With the same intermittent triglyceride, the OD of the groups with high concentration of glucose were lower than the groups with low concentration of glucose.2 The standard of CRP:With the same concentration of glucose, the content of inflammatory CRP in the groups with sustained abnomal concentration of triglyceride and the groups with triglyceride fluctuation were higher than the groups with low concentration of triglyceride. With low concentration of glucose, CRP in the groups with middle concentration of TG had no significant difference with high concentration of TG. The group with high glucose joint training, compared with the group in low concentration of triglyceride, There was no significant difference content of PCR in Intermittend triglyceride groups. The content of ICAM in the group with high concentration of triglyceride and high concentration of glucose was the highest. With the same concentration of triglyceride, the CRP of the groups with high concentration of glucose were higher than the groups with low concentration of triglyceride. The CRP of onefold high concentration glucose groups were lower than the groups with onefold high triglyceride. While the CRP of the group with high concentration of triglyceride and high concentration of glucose joint was the highest. With the same intermittent triglyceride, the CRP of the groups with high concentration of glucose were higher than the groups with low concentration of glucose.3 The standard of ICAM:With the same concentration of glucose, the content of inflammatory ICAM in the groups with sustained abnomal concentration of triglyceride and the groups with triglyceride fluctuation were higher than the groups with low concentration of triglyceride. With low concentration of glucose, ICAM in the groups were higher then the group with low concentration of triglyceride except the group with middle fluctuation of TG. The group with high glucose joint training, compared with the group in low concentration of triglyceride, There was no significant difference content of ICAM in Intermittend triglyceride groups. The content of ICAM in the group with high concentration of triglyceride and high concentration of glucose was the lowest. With the same concentration of triglyceride,the ICAM of the groups with high concentration of glucose were higher than the groups with low concentration of triglyceride. The ICAM of onefold high concentration glucose groups were lower than the groups with onefold high triglyceride. While the ICAM of the group with high concentration of triglyceride and high concentration of glucose joint was the highest. With the same intermittent triglyceride, the ICAM of the groups with high concentration of glucose were higher than the groups with low concentration of glucose.4 The standard of VCAM:With the same concentration of glucose, the content of inflammatory VCAM in the groups with sustained abnomal concentration of triglyceride and the groups with triglyceride fluctuation were higher than the group with low concentration of triglyceride. With low concentration of glucose, VCAM in the groups of intermittend high concentrition triglyceride or sustained high concentrition triglyceride were significantly higher than the group with low concentration of triglyceride, but the other groups were not significantly higher. The group with high glucose joint training, compared with the group in low concentration of triglyceride, there was no significant difference content of VCAM in intermittend triglyceride groups, but the other groups were significantly higher. With the same concentration of triglyceride, the VCAM of the groups with high concentration of glucose were higher than the groups with low concentration of triglyceride. The VCAM of onefold high concentration glucose groups were lower than the groups with onefold high triglyceride. While the VCAM of the group with high concentration of triglyceride and high concentration of glucose joint was the highest. With the same intermittent triglyceride, the VCAM of the groups with high concentration of glucose were higher than the groups with low concentration of glucose.5 The standard of MCP-1:With the same concentration of glucose, the content of inflammatory MCP-1 in the groups with sustained abnomal concentration of triglyceride and the groups with triglyceride fluctuation were higher than the group with low concentration of triglyceride. MCP-1 in the groups of intermittend high concentrition triglyceride or sustained high concentrition triglyceride were higher than the group with low concentration of triglyceride and the other groups were not. The groups with high glucose joint training, compared with the group in low concentration of triglyceride, the groups with sustained high concentration of triglyceride were significantly higher, but there was not significant difference content of MCP-1 compared with other groups. With the same concentration of triglyceride, the MCP-1 of the groups with high concentration of glucose were higher than the groups with low concentration of triglyceride. The MCP-1 of onefold high concentration groups were lower than the groups with onefold high triglyceride. While the MCP-1 of the group with high concentration glucose of triglyceride and high concentration of glucose joint was the highest. With the same intermittent triglyceride, the MCP-1 of the groups with high concentration of glucose were higher than the groups with low concentration of glucose.6 The standard of MDA:With the same concentration of glucose, the content of MDA, which is the production from inflammation,in the groups with sustained abnomal concentration of triglyceride and the groups with triglyceride fluctuation was higher than the group with low concentration of triglyceride. But the difference betwen groups with triglyceride fluctuation and the group with low concentration of triglyceride was not statistically significant. The content of MDA in the group with high concentration of triglyceride and high concentration of glucose was the highest. With the same concentration of triglyceride,the VMDA of the groups with high concentration of glucose were higher than the groups with low concentration of triglyceride. The MDA of onefold high concentration glucose groups were lower than the groups with onefold high triglyceride. While the MDA of the group with high concentration of triglyceride and high concentration of glucose joint was the highest. With the same intermittent triglyceride, the MDA of the groups with high concentration of glucose were higher than the groups with low concentration of glucose.7 The standard of SOD and MDA:With the same concentration of glucose, the content of SOD and NO,which can protect the vascular from infiammation,in the groups with sustained abnomal concentration of triglyceride and the groups with triglyceride fluctuation was lowwer than the group with low concentration of triglyceride.But the content of the groups with triglyceride fluctuation were not lowwer than the group with sustained low triglyceride. With the same concentration of triglyceride, the of the groups with high concentration of glucose were lower than the groups with low concentration of triglyceride. The SOD and NO of onefold high concentration glucose groups were higher than the groups with onefold high triglyceride. While the SOD and NO of the group with high concentration of triglyceride and high concentration of glucose joint was the lowest. With the same intermittent triglyceride, the SOD and MDA of the groups with high concentration of glucose were lower than the groups with low concentration of glucose.Conclusion:1 With the same concentration of glucose,intermittend triglyceride and sustained middle concentration triglyceride can promote cell proliferation in both, but low concentration of glucose as well as middle concentration of triglyceride was the most effective.Sustained high triglyceride concentration inhibited cell proliferation, high triglyceride and high glucose joint training was the most effective. The ability of inhibited cell proliferation of onefold high concentration triglyceride groups were higher than the groups with onefold high glucose.2 Intermittend triglyceride and sustained hypertriglyceride damaged HUVEC,but the damage of intermittend triglyceride was not larger. The ability of damaging HUVEC of onefold high triglyceride were higher than onefold high glucose, while high triglyceride and high glucose joint training was the most effective. With the same intermittent triglyceride, the ability of damaging HUVEC of high glucose training was higher than the low glucose.3 The antioxidative ability was weakened with intermittent triglyceride or sustained hypertriglyceride,but the weakness of the former is not larger then the latter The ability of antioxidation of onefold high concentration triglyceride groups were lower than the groups with onefold high glucose, while highest triglyceride in this experiment and high glucose joint training was the most effective. With the same intermittent triglyceride, the ability of antioxidation of HUVEC high glucose training were higher than low glucose.4 The function of the HUVEC could be damaged by Intermittend triglyceride or sustained hypertriglyceride( decreased NO content); but the effect of the former is not larger then the latter. The ability of damaging the function of HUVEC of onefold high triglyceride groups were higher than onefold high glucose, while high triglyceride and high glucose joint training was the most effective. With the same intermittent triglyceride, the ability of damaging the function of HUVEC of high glucose training was higher than the low glucose.
