Purpose:The present study explored the methylation status of gene RASSF1A andgene ZO-1promoter region in the bone marrow of acute myeloid leukemia (AML) patientswith different syndromes. The study provides more evidence to acute leukemia diagnosisand treatment by investigating the role methylation status of the two genes plays indifferent syndromes.Method:41bone marrow samples of different AML subtypes andillness time were collected. They were divided into subgroups by traditional Chinesemedical (TCM) diagnostic criteria:18samples for deficiency of both vital energy and yin,8samples for intense toxic heat, and15for Blood stasis phlegm knot. For the controlgroup,10bone marrow samples and15peripheral blood samples of healthy people werecollected, and mononuclear cell was extracted. The methylation status of gene RASSF1Aand gene ZO-1promoter region was tested using methylation specific polymerease chainreaction (MSP-PCR). The results were analyzed and interpreted through the viewpoint ofmolecular biology. Result:(1)There was no significant difference in the methylationstatus of gene RASSF1A and gene ZO-1promoter region among the three subtypes: thedeficiency of both vital energy and yin, intense toxic heat, and blood stasis phlegm knot(P>0.05).(2) Subtypes of AML patients M1, M2, M3, M4and M5all showed high level ofmethylation. There was no significant difference in methylation positive rate between thesubtypes (P>0.05).(3) There was no significant difference in methylation positive ratebetween early patients and relapsing patients.(4) Patients with positive methylation ratehad significant high level of peripheral blood leucocyte (P<0.05) and significant high level of myeloid progenitor cell.(5) of the41AML patients,22showed RASSF1A methylation(positive), and20showed ZO-1methylation (positive). All were significantly differentfrom the control group (P<0.05).Conclusion:Empirical patients show higher level ofmethylation rate in gene RASSF1A and gene ZO-1than deficient patients. The methylationpositive rate from molecular level indicates the essence of TCM real memory;RASSF1Aand ZO-1gene showed hypermethy lation of expression in leukemia cells with white bloodcell count,high proportion of bone marrow blast cells in the clinicalcharacteristics,suggesting that RASSF1Aand ZO-1gene play a role in the pathogenesis ofAML,Contribute to the diagnosis and targeted therapy of AML.
