Objective: Aim to extract and purify the Oldenlandia diffusa flavonoids andconduct a research on its Intestinal flora Metabolism. Method: The singlefactor investigation combined with orthogonal test were used for screeningthe extraction technology of Oldenlandia diffusa flavonoids, the AB-8macroporous adsorption resin and the polyamide resin were used for flavonoidspurification and flavonoid glycoside purification; use acid hydrolysis forOldenlandia diffusa flavonoid aglycon production; flavonoid glycoside andflavonoid aglycon’s intestinal absorption were compared by using rats in vivointestinal perfusion test and the intestinal eversion test;observe andcompare the Intestinal flora Metabolism of rutin and quercetin in rat and human,using fresh feces at37℃and rats anaerobic culture, and analyze themetabolin. Results: Use70%ethanol for reflux extraction of total flavonoidsand AB-8macroporous resin for concentration of active ingredients,and atthe same time, to remove impurities and improve the purity of flavonoid;polyamide adsorption resin could purify the Oldenlandia diffusa flavonoidglycoside effectively; the acid hydrolysis was a simple method to getflavonoid aglycon with a high yield; under in vitro conditions, the rutin andquercetin in Oldenlandia diffusa could be transformed to aglycone in the ratand human intestinal flora, and the human intestinal flora work more quicklycompared with the rat’s. Conclusion: Oldenlandia diffusa flavonoid glycoside appears to work by generating aglycone or Secondary glycosides with greateractivity or higher bioavailability through Intestinal flora Metabolism.
