The Preliminary Study Of Anti-tumor Effect Of The Extract Of Oldenlandia Diffusa

Objective: To investigate the anti-tumor effect and mechanism of the extract of Oldenlandia diffusa (OD).Methods: Horn method was used to calculate mice acute oral LD50 values (withtheir 95 % confident limits). OD extract was administered orally to mice, and tumor inhibition action on S180 sarcoma-bearing mice was observed. Anti-proliferation effect of the OD extract to the K562 leukaemic cell line in vivo by MTT cytotoxic assay and to Hela cell line in vivo by seropharmacological method and MTT assay. Superoxide dismutase (SOD) and malondialdehyde (MDA) were detected by colorimetry.Results: 1. Mice acute oral LD50 values (with their 95 % confident limits) of OD extract were 794mg/kg (489-1290mg/kg) in female and 909mg/kg (546-1510mg/kg) in male. 2. In the group of high, middle and low dosage, the tumor inhibition rate on mice-bearing S180 were 32.23%, 34.40% and 35.54% respectively, OD extract inhibited the growth of solid tumor of mice-bearing S180 obviously (compared with model group low dosage group P < 0.05, middle and low dosage group P < 0.01). 3. The suppress rate of K562 cells treated with OD extract that with concentration of 10 μ g/ml, 40 μ g/ml and 80 μ g/ml were 53.12%, 82.74% and 98.27% respectively, suppressed proliferation obviously (P < 0.01). 4. The suppress rate of Hela cells treated with OD extract-containing rat serum that were inactivated and with concentration of 10%, 20% and 25% were 35.72%, 33.73% and 36.01% respectively, and were 36.01%, 38.85%, 48.24% respectively treated with OD extract-containing rat serum that were not inactivated, showing more stable suppression and dosagedependent effect, all suppressed proliferation obviously (P < 0.01). 5. The activity of SOD was lower and the content of MDA was higher in model group than that in control group ( P < 0.01) . while The activity of SOD was higher and the content of MDA was lower in OD extract treated group than that in control group and approach to the levels of control group ( P < 0.01).Conclusion: 1. OD extract have the effect of inhibiting the growth of S180 sarcoma in vivo and anti-proliferation effect for K562 cell line in vitro. 2. OD extract-containing serum have the effect of inhibiting proliferation of Hela cell line. 3. OD extract have antioxidant effect that is one of its anti-tumor mechanisms. 4. In the results of acute oral toxicity study test, the OD extract belong to low toxicity Chinese medicine.
