Objective:To investigate the feasibility of gene therapy for meningioma by theChemically synthesized of small interfering RNA (siRNA) to inhibit HER2geneexpression in vitro.Methods: Cells from fresh and sterile meningioma tissue were cultured、passaged and depurated by trypsin digestion. Select the meningioma cells of HER2protein overexpression which identified by immunocytochemistry. SiRNA targetingHER2gene was designed and synthesized, set up of blank control group, emptyvector group, non-specific siRNA group, and HER2/neu siRNA group. SiRNA wastransfected into meningioma cells using cationic liposome Lipofectamine○R2000astransfecing agent,The HER2and Ki-67protein expression in meningioma cells wereexamined by immunocytochemistry methods in48hours after transfection,.Results:The meningioma cells were cultured successfully, and identified byimmunocytochemistry methods for EMA、Vimetin and HER2. Immunocytochemistryexperimental results showed: compared with blank control group、empty vector group,non-specific siRNA group,there were significant decrease of the HER2and Ki-67protein expression in the HER2siRNA group(P<0.01).In contrast,there were noobvious changes in blank control group, empty vector group and non-specificsiRNA group(P>0.05).We also investigated that the expression of HER2wascorrelated with Ki-67through test of Spearman’s rank-correlation coefficient(rs=0.616,P<0.001).Conclusions:(1)The human meningioma cells were cultured successfully, wesuggest that meningioma cells experiments for RNAi and drug in vitro should start inthe third generation.(2).RNAi mediated by chemically synthesized of specific siRNAcan effectively inhibit the expression of HER2protein in human meningioma cells,(3)The cellular proliferation and Ki-67protein significantly decreased in meningiomacells after inhibited the expression of HER2,so RNAi may be a novel method to treatmeningiomas which HER2protein overexpression....
