Background:Functional dyspepsia (FD) is a common problem affecting up to10-25%of individuals.FD accounts for significant health care costs and affects quality of life but has no definitive pathophysiology and treatment. Exogenous GLP-1slows gastric emptying, suppress appetite, inhibit small intestinal motility in healthy, obese, type2diabetic,and critically ill subjects.however, the effects of GLP-1have not been evaluated in FD patients.Purpose.This study was designed to test whether fasted serum GLP-1concentration is variation in FD patients by comparison with healthy controls,and if so,to further investigating the fasted serum GLP-1concentration whether affected by different clinical subtype and severity scales of FD. Try to supplement pathophysiology of FD and investigate potential treatment for FD.Methods:1.Object of study:From December2011to May2012,50consecutive outpatients with FD from our department were enrolled according to the Rome III criteria.All recruited cases were subdivided into postprandial distress syndrome(PDS),27cases,epigastric pain syndrome(EPS),27cases; Additional, according to dyspepsia symptoms severity self-report scale,they were divide into three groups,mild9cases, moderate19cases, severe22cases,and20healthy controls were enrolled in this study. Comparing the fasted serum GLP-1concentration between each groups.2.Determination fasted serum levels of GLP-1of the experimental group and control group of FD patients:extract2ml blood from all FD patients and healthy volunteers since morning on an empty stomach, after high speed centrifugation to separate serum in EP tubes,-80℃refrigerator save, enzyme linked immunosorbent assay kit (ELISA) for measuring the concentration of the serum free GLP-1.3.Statistical processing:the data as a result, the normal distribution with mean±standard deviation (x±s)non-normal distribution in median fire interquartile range (M25-75%); Using Pearson correlation analysis (normal distribution) or Spearman (non normal distribution) test. All data using SPSS13.0statistical software for statistical analysis. Comparing FD patients and healthy volunteers,FD patients two subtypes and two symptoms integral group, serum levels of GLP-1levels.comparison between the two groups using/test. All inspection are used on both sides, with P<0.05think difference is statistically significant.Results:1.FD group and healthy volunteers GLP-1level change and comparison FD group and healthy group average serum levels of GLP-1levels were87.04±40.17pg/ml,153.55±21.76pg/ml. FD group compared with healthy group, serum levels of GLP-1levels was statistically significant (P=0.003,<0.05).2.EPS and PDS group changes in serum levels of GLP-1and comparison EPS and PDS group average serum levels of GLP-1levels were89.78±42.65pg/ml,83.67±37.87pg/ml. EPS and PDS group compare serum levels of GLP-1has no obvious statistical significance (P=0.787,>0.05)3.Mild to severe symptoms integral set of changes in serum levels of GLP-1and comparison According to the mild, moderate group and severe group GLP-1levels were83.31±32.67pg/ml,86.59±32.67pg/ml and32.67±39.78pg/ml. Mild group and severe group, serum levels of GLP-1levels are no significant statistical significance (P=0.494,>0.05). Mild group and moderate comparing serum levels of GLP-1has no obvious statistical significance (P=0.536,>0.05). Serum levels of GLP-1moderate group and severe group is no significant statistical significance (P=0.534,>0.05).Conclusion:1.The fasted serum levels of GLP-1of FD patients compared with healthy subjects decreased significantly, the difference was statistically significant. GLP-1may participate in the pathogenesis of FD. 2.FD two clinical subtypes in patients with fasted serum levels of GLP-1level difference is not significant. FD patients the levels of fasted serum levels of GLP-1.3.FD indigestion symptom severity integral light, medium and severe patients, fasting serum GLP-1level difference is not significant. FD patients, fasting serum levels of GLP-1levels are not influenced by patients self-conscious symptom severity.
