Objective:Study the application of a-chymotrypsin in the processing of high viscosity semen. Materials&methods:According to the drawing,128fresh semen were divided into three groups:normal consistency semen49, medium viscose26, higher viscose semen53, each sperm was divided into two,one is treated with a-chymotrypsin another without a-chymotrypsin, two sperm were processed with Percoll density gradient centrifugation,caculate the recovery rate of processive sperm about the two processing in different viscosity group. At the same time, work out the rate of sperm with normal morphology and DFI of fresh semen, sperm suspension after using a-chymotrypsin or not a-chymotrypsin, analyze the related parameters using SPSS. Results:1) For normal and medium viscosity semen group, there was no significant difference about the processive sperm recovery rate with or with not a-chymotrypsin. For high viscosity group, there was significant difference about the processive sperm recovery rate;2) there was no statistical difference between with and with not a-chymotrypsin about the rate of sperm with normal morphology and DFI;3) there was statistical difference between fresh semen and sperm after using a-chymotrypsin or not using a-chymotrypsin;ConcIusion:1) a-chymotrypsin can significantly improve the processive sperm recovery rate during percoll density gradient centrifugation, there was no bad effect on normal morphology and DNA integrity of sperm.2) Percoll density gradient centrifugation can improve the percentage of sperm with normal morphology and DNA integrity. Chapter2Abstract:The clinical application value of CAS A parameters in IUI and AID.Objective:1) Predictive value of CAS A parameters in AID with froze donor semen;2) Predictive value of CASA parameters in IUI with husband fresh semen. Materials&methods:According to certain requirements, we collected clinical data of145patients who accepted IUI treatment and122patients who accepted AID treatment from September2012to January2013in our hospital, semen analysis was performed using CASA, and SPSS was used to analyze the differences of CASA parameters between pregnant and non-pregnant in IUI and AID. Results:1) outcome of IUI:there was statistical difference about (A+B)%、VSL、 VAP、VCL before processing and sperm concentration VCL、ALH after processing between pregnant and non-pregnant. The AUC is respectively0.651、0.656、0.647、0.649、0.655、0.707、0.776, there was statistical difference about (A+B)%、VAP、VSL、VCL、ALH of frozen donor semen after thawing between pregnant and non-pregnant. The AUC is respectively0.618、0.633、0.610、0.633、0.629.Conclusion:Compared with other parameters, VCL、ALH after processing seems to be more useful to predict pregnancy outcome of IUI;(A+B)%、VAP、VCL、 VSL、ALH of frozen semen after thawing are associated with pregnancy outcome, but the predictive value is not high.
