IFN-γ is generated by NK cells and activated T cells (including CD8+T cells and ThO cells, Thl cells).IFN-γ can interact with extracellular matrix cooperative form, in order to paracrine manner to the corresponding cells. Its species specificities include antiviral, antiproliferative, it also can regulate immune functions,these characteristics make it an important factor in infectious diseases progressions. Especially in the HIV/AIDS patients with HAART, it plays a key role. This paper observed the effect of IFN-γ on highly active one year antiretroviral therapy (highly active antiretroviral therapy, HAART) to HIV/AIDS pations.Objective:To investigate the effect of IFN-y to HIV/AIDS patients on one-year HAART.Methods:20HIV/AIDS patients treated with HAART were selected randomly, who were compared with20healthy subjects.10ml Blood samples were taken at the time point0,6,12months. Then PBMCs was aquired in the next4hours and would be cultured for5days with or without IFN-y.5days later collect the supematants and substrate, then test IL-12p70and IP-10with ELISA, CD4+CD25+Foxp3Treg cells with cell flow.Results:The CD4+T cell count (X±SD) of the20HIV/AIDS patients in group were124.7±86.4/μl,203.8±98.3/μl,376.2±142/μl in the time of pre-HAART,6months,12months, Statistics show that CD4+T lymphocytes increased significantly as the HAART treatment prolonged (P<0.05). The results of IL-12level in supernatant (simple/IFN-γ co-culture) respectively were [(37.02±12.76) vs (41.79±15.02),t=2.336, P=0.03]、[(41.76±17.01) vs (47.2±14.26), t=2.702, P=0.014]、[(48.01±11.84) vs(53.44±11.3),t=3.14, P=0.003], P<0.05, there were statistical significance. The results of IP-10level respectively were [(175.7±53.17)vs(231.32±73.08), t=1.68, P=0.109]、[(117.8±65.12) vs(174.01±85.77),t=2.26,P=0.03]、[(76.87±45.56)vs(102.84±62.48), t=3.43, P=0.002], no statistically significant was found in HAART0month group (P>0.05), there is statistical significance in HAART6,12months(P<0.05). CD4+CD25+FOXP3Treg cell percentage were tested by flow cytometry,and the results respect were [(10.41±1.1) vs (2.4±1.11)%, t=13.89, P=0.000]、[(8.33±2.03) vs (1.99±0.86)%, t=12.93, P=0.000]、[(5.65±1.55) vs (1.32±0.73)%, t=10.61, P=0.000], P<0.05, there were statistical significance. There were positive correlation between IL-12P70/IP-10and HIV-RNA.Conclusion:(1) With HAART treatment time prolonged, CD4+Tcells were increased, CD4+CD25+FOXP3Treg cells percentage were decreased;(2) HAART treatment increased IL-12P70concentration, and decreased the concentration of IP-10;(3) IFN-γ can stimulate secretion of IL-12P70and IP-10,to help restore the immune function.20 charts,11tables,79references...
