After HIV infected human body,it infringes upon the body’s immune system, causes the immune function lacking collapse and ultimately leads to AIDS.Through suppressing HIV virus replication,HARRT can recover immune function of patients to some extent. This paper observes one-year HAART on immune reconstruction and corresponding cell factors in the HIV/AIDS patients. This study includes two parts as follows:PART Ⅰ The effect of one-year highly active antiretroviral therapy on HIV/AIDS immune reconstructionObjective:To investigate the impact of one-year HAART on antiviral effect and immune reconstruction in the HIV/AIDS patientsMethods:35HIV/AIDS patients treated with HAART were selected randomly, who were compared with35healthy subjects. Blood samples were taken at the time point0,6,12months. The virus vector of the objects and the change of T cell subsets were under surveillance.Results:The virus vector of35patients treated with half-year HAART reduced to below detected values. Moreover, although the level of CD4+virginity cells increased by8个/μl (P>0.05)、77个/μl(P <0.01) compared with the baseline after they were treated6months and12months, respectively, it were still lower than healthy control group; the level of CD4+memory cells increased by57个/μl (P<0.01)、93个/μl (P<0.01) compared with the baseline after they were treated6months and12months, respectively, it were still lower than healthy control group; the level of CD8+activated cells decreased by122个/ul (P<0.01)、170个/μl (P<0.01) compared with the baseline after they were treated6months and12months, respectively, it were still lower than healthy control group.Conclusion:HAART not only can inhibit virus replication rapidly, but also can increase CD4+virginity cells and CD4+memory cells and decrease CD8+activated cells gradually, so that it contributes to the immune reconstruction for HIV/AIDS patients.PART ⅡThe change of IL-12、IFN-γ and IP-10in HIV/AIDS serum with one-year HAART and its relationship with immune reconstructionObjective:To investigate the dynamic changes of IL-12、IFN-γ and IP-10in HIV/AIDS patients in China after one-year HAART and analyze the relationship and its significance between above three factors and the main immune reconstruction and immune activation indexMethods:35HIV/AIDS patients treated with HAART were selected randomly, who were compared with35healthy subjects. Blood samples were taken at the time point0,6,12months. The change of IL-12、IFN-γ and IP-10of the objects were under surveillance and their relationship with T cell subsets were analyzed.Results:The level of IL-12, IFN-γ and IP-10before and after treatment of6months and12months with HAART in the serum of HIV/AIDS patients and normal subjects were listed as follows:IL-12:(25.24±13.88) pg/ml、(25.27±10.57) pg/ml、(29.91±8.57) pg/ml、(33.15±8.32) pg/ml、IFN-γ:(27.84±13.06) pg/ml、(28.81±11.96) pg/ml、(32.38±2.37) pg/ml、(34.55±13.67) pg/ml,IP-10:(429±172.52)pg/ml、(320±156.95)pg/ml、(236±120.67) pg/ml、(90.29±38.06) pg/ml、The level of IL-12in serum was in a positive relationship with CD4+virginity cells and CD4+memory cells.It showed no obvious relationship with CD8+activated cells (CD8+CD38+); the level of IFN-γ showed positive correlation with the level of IL-12, and had no obvious relationship with CD8+T lymphocytes; the level of IP-10had positive relationship with CD8+activated cells (CD8+CD38+), and had negative relationship with CD4+T lymphocytes.Conclusion:The incidence of HIV infection could lead to parasecretion of IL-12、IFN-γ and IP-10. HAART could correct its abnormal level to some extent. Moreover, IL-12、IFN-γ and IP-10could be used as related cytokines for immune reconstruction and immune activation and also effective criterion for curative efficacy of HAART.
