The Expression And Significance Of TNF-a, JNK, C-jun In Placental Tissue Of Patients With Intrahepatic Cholestasis Of Pregnancy

Objective:To investigate the expression and significance of tumor necrosis factor-alpha (TNF-alpha), JNK, c-jun in placental tissue of patients with intrahepatic cholestasis of pregnancy.Methods:detect the expressions of TNF-alpha, JNK, c-jun in the placental tissue of25cases of ICP (experimental group) and25normal pregnant women (control group) in our hospital by Immunohistochemical SP method,the degree of the expression level integrated optical density,that IOD. using the t test to compare the expressions between ICP group and control group. The25cases of ICP were divided into ICP mild group (17cases) and ICP severe group (8cases), then using analysis of variance for ICP mild group, ICP severe group and normal control group. Using LSD-t test for multiple comparisons between the three groups if Analysis of variance was statistically significant, correlation analysis between TNF-α JNK、c-jun.Results:(1) t-test between ICP group and the control group:the expressions of TNF-a, JNK, c-jun in placental tissue of ICP group were higher than the normal control group, and statistically significant.(2) analysis of variance and multiple comparisons among ICP mild group, ICP severe group and normal control group:TNF-a, JNK, c-jun in the three groups were measured by analysis of variance P<0.05, are statistically significant, further multiple comparisons among the three groups. using LSD-t test to compare the expression of TNF-a between ICP mild group and severe group, P>0.05, which is not statistically significant; using LSD-t test to compare the expression of TNF-a In ICP mild group and the normal control group, ICP severe group and the normal control group, P<0.05, both are statistically significant, using LSD-t test to compare the expression of JNK In ICP mild group and the normal control group, ICP severe group and the normal control group, ICP mild group and ICP severe group,P<0.05, three are statistically significant.(3) correlation analysis between TNF-α、JNK、c-jun: There is no obvious straight line relationship between TNF-alpha and JNK, either between TNF-alpha and c-jun. There is a linear relationship between JNK and c-jun, the Pearson correlation coefficient is0.789, and P<0.01, which was statistically significant.Conclusion:the high expression of TNF-alpha, JNK, c-jun in placental tissue of patients with intrahepatic cholestasis of pregnancy may be play a role in the pathogenesis of ICP. If the degree of the expression of the three affect the severity of ICP, and if there is interaction between JNK and c-jun to affect the occurrence of ICP, these deserve further study.
