Purpose:Studies on the preparation process of Salvianolic acid A.Material and method:1.According to the single and orthogonal design,the extraction process of Salvianolic acid B from Alvia miltiorrhiza Bge using extraction rate as target indicator.By optimizing the condition of macroporous adsorption resin column chromatography processes,the optimum condition of separation was determined.2.Based on the degradation mechanism from Salvianolic acid B to Salvianolic acid A,the different parameters affecting the transformation effects were investigated by the single and orthogonal design.3.This research studies the separation and purification process of Salvianolic acid A by macroporous adsorption resin colunm chromatography,sephadex column chromatography and reversed phase chromatographic packing column chromatography.Results:1.It was found that 1:10 water to Salvia miltiorrhiza the weigh ratio was used last for 2 h twice at the temperature of 70℃.The average extraction rate was above 90%.The results of experiment indicate that the HPD100 macroporous adsorptive resin has best adsorption and desorption effect on the Salvanobic acid B.2.The best transforming process were found as follow:the concentration of Salvianolic acid B is 15 mg/mL;pH value,5;temperature,120℃;time of transformation,4 h.3.The optimum adsorption and elution conditions of column chromatography were found. The purity of Salvianolic acid A was as high as 94%.4.Through chemical compositions study of this extract,we got components through column chromatography and identified the structures according to physical-chemical properties and spectrum data.They are Danshensu,protocatechualdehyde,rosmarinic acid,lithospermic acid, salvianolic acid B. Conclusion:We obtained a simple,efficient and reasonable route of technics.It will provide a beneficial reference for the development of salvianolic acid A.
