Diabetes is a chronic disease and can’t be cured by now; the need is to be lifelong treatment. Therefore, the discovery of new anti-diabetic drugs is a key field in the research and development of new drugs. In our previous study, the common120heat-clearing herbs have been screened for the hypoglycemic activity finding that traditional Chinese medicine Ampelopsis japonica has good hypoglycemic activity. Its crude extract was extracted with petroleum ether, ethyl acetate and n-butanol getting three fractions, the ethyl acetate fraction of which showed greatest hypoglycemic activity.Based on the previous study, this research used a-amylase and DPP-4model in vitro to screen the activity parts of Ampelopsis japonica (Thunb.) Makino, the separation and purification of the active ethyl acetate fraction was obtained by variety of chromatographic separation technologies.Forty-four compounds were isolated and identified as b1-1:β-sitosterol lin-oleate, b1-2:linoleate, b1-3:β-sitosterol, b1-4:paeonol, b1-5:lupeone, b1-6:a-to-copherol, b1-7:physcion, bl-8:bis(2-ethylhexyl)phthalate, b1-9:24?-n-propyl ch-olestan-3-one, b1-10:β-sitosterol linolenate, b1-11:1,2,3-trilinolein, b1-12:lupeol, b1-13:a-linolenic acid, b1-14:a-tocopherol quinone, bl-15:a-methyl pyrrole ket-one, b1-16:1-acetyl-β-carboline, bl-17:betulin, b1-18:vanillin, b1-19:undecano-ic acid, b1-20:5a,8a-epidioxyergosta-6,22-dien-3β-ol, b1-21:botcinins D, b1-22: lasiodiplodin, b1-23:4-keto-pinoresinol, bl-24:rel-(3R,3’S,4R,4’S)-3,3’,4,4’-Tetrah-ydro-6,6’-dimethoxy[3.3’-bi-2H-benzopyran]-4,4’-diol, b1-25:(+)-epipinoresinol, b1-26:3,5-dimethoxy-4-hedroxyphenyl-propynol, b1-27:methyl3a-trans-feruloyloxy-2a-hydroxyurs-12-en-28-oate, bl-28:(6R,7E,9S)-9-hydroxy-4,7-megastigmadien-3-one, b1-29:S-(+)-dehydrovomifoliol, b1-30:2-cyclohexen-1-one,4-(5-hydroxy-3-m-ethyl-1,3-pentadienyl)-3,5,5-trimethyl, bl-31:3a-trans-feruloyloxy-2a-O-acetylurs-12-en-28-oic acid, b1-32:3-furoic acid, bl-33:vanillic acid, b1-34:(7S,8S)-3-me -thoxy-3’,7-epoxy-8,4’-oxyneoligna-4,9,9’-triol, b1-35:(2S)-1,2,6’-tri-O-[(9Z,12Z,15Z)-octadeca-9,12,15-trienoyl]-3-O-β-D-galactopyranosyl-glycero, b1-36:(+)-absc-isin-Ⅱ, b1-37:linoleic acid-1-glyceride, bl-38:methyl pyroglutamate, b1-39:eu-scaphic acid, b1-40:3,4-ethylparaben, b1-41:4,5-dihydroblumenolA, b1-42:4-p-menthane-1,8-diol, b1-43:4-ethyl-2-hydroxy-succinate, b1-44:bungeinA.These compounds include5sesquiterpeness and nor-sesquiterpenes,5phenylpropanoids,7triterpenoids,4sterols,6fatty acids and glycerol esters and17other types compounds. Compounds b1-1, b1-2, b1-4-b1-6, bl-8-b1-11, b1-13-b1-26, bl-28-b1-30, b1-32-b1-44were obtained from Ampelopsis japonica (Thunb.) Makino for the first time. Compounds b1-27, bl-31were two new triterpenoids isolated from this plant. Compounds b1-5, b1-6, b1-9, b1-12, b1-16, b1-17, b1-20, b1-21, b1-22,b1-23, b1-25, b1-27, b1-31, b1-34, b1-39showed some hypoglycemic activity.
