Objective:To observe the influence of mixture of Maxingtingxiong dose on hemodynamics and right heart function of model rats and exploring its mechanism.Methods:50SD rats were sex-randomly divided into normal control groups, model groups, Fasudil group, large-dosage Chinese herb group and mid-dosage Chinese herb group. Each group of10rats contains half male and half female.Useing normal pressure and hypoxic environment and simulating rats model of HPH,they have been given21days of drug intervention.Right heart catheterization measures the mean pulmonary arterial pressure and right ventricular systolic pressure.The left common carotid arterial intubation measure the mean carotid artery pressure. Radioimmunoassay determines of AT-Ⅱ content in rat plasma.ELISA determines the5-HT and NT-proBNP content in rat plasma. Weighing the right ventricle (RV), left ventricle (LV)and septal (S), calculate the degree of right ventricular hypertrophy by right ventricular hypertrophy index [RV/(LV+S)].Results:(1) The values of mPAP (mmHg, x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group and Fasudil group are21.61±0.38,23.25±1.19,18.85±0.65, which are all lower than that of model groups30.27±1.47, the difference was statistically significant (P<0.05); The values of mCAP (mmHg, x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group,Fasudil group and model groups are112.31±2.16,113.33±1.54,112.09±1.82,112.69±3.71, compared with the model groups are not significantly different(P>0.05).(2) The values of RVHI%(x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group and Fasudil group are28.19±1.67,27.64±1.55,30.24±1.82, which are all lower than that of model groups41.41±3.71,the difference was statistically significant(P<0.05); The values of RVSP (mmHg, x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group and Fasudil group are29.56±0.79,29.82±1.73,26.74±1.63, which are all lower than that of model groups35.25±1.43,the difference was statistically significant(P<0.05); The values of NT-proBNP (fmol/ml,x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group and Fasudil group are192.31±4.59,196.25±2.73,165.93±3.76, which are all lower than that of model groups246.51±3.34,the difference was statistically significant(P<0.05).(3) The values of5-HT (μg/1, x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group and Fasudil group are30.46±5.46,29.15±4.56,31.25±4.31, which are all lower than that of model groups85.25±6.35,the difference was statistically significant(P<0.05); The values of AT-Ⅱ(pg/ml,x±s) of mid-dosage Chinese herb group,large-dosage Chinese herb group and Fasudil group are308.25±25.47,305.17±24.35,311.27±21.20, which are all lower than that of model groups482.38±20.21,the difference was statistically significant(P<0.05).Conclusions:(1) Mixture of Maxingtingxiong can effectively reduce the pulmonary artery mean pressure in Hypoxic pulmonary hypertension rat, without affecting the systemic circulation. We can conclude that mixture of Maxingtingxiong plays an important role in preventing pulmonary hypertension.(2) Mixture of Maxingtingxiong can effectively reduce the right ventricular hypertrophy in Hypoxic pulmonary hypertension rat.mixture of Maxingtingxiong can influence the occurrence and development of right heart failure in Hypoxic pulmonary hypertension rat.(3) Changes of5-HT and AT-Ⅱ content in rat plasma plays an important role in preventing pulmonary hypertension. Mixture of Maxingtingxiong can reduce5-HT levels and AT-Ⅱ levels of hypoxic pulmonary hypertension in rat’s plasma, which may be one of the mechanisms that Mixture of Maxingtingxiong to influence the occurrence and development of preventing pulmonary hypertension and right heart failure.(4)According to traditional Chinese medicine theory,Mixture of Maxingtingxiong composition method and pharmacological analysis can confirmed the Mixture of Maxingtingxiong have releasing lung and removing blood stasis and resolving phlegm effect.Mixture of Maxingtingxiong have the exact effect of traditional Chinese medicine dialectical treatment of HPH.
