| Objective To survey the effection of low tempreture on platelet apoptosis.Methods Washed healthy people platelets were stored at0℃,4℃,10℃,22℃and37℃for up to5days. All samples were measured every day.Flow cytometrydetected the change of mitochondrial membrane potential. Western blot analysis theapoptosis proteins caspase-3and antiapoptosis proteins Bcl-xl express.Results The data from flow cytometry indicate that every group exists thechange of depolarization of mitochondrial transmembrane potential (△Ψm).Compared with other groups, the depolarizations of△Ψm of the group which wasstored at10℃were inhibited significantly.When stored at10℃,several apoptoicevents in apoptosis including down-regulation of Bcl-xl and caspase-3activation wereinhibited significantly, according to the results of western blot.However, when plateletsstored at the other groups, apoptosis protein caspase-3were activated and expressionof anti-apoptosis protein Bcl-xl were decreased.Conclusion These data suggest that low tempreture inhibits platelet apoptosis.The experimental results suggest that attention should be paid to platelet apoptosisunder low tempreture conditions such as during platelet storage. |
