| Objective:Studying the probable repair mechanism of moxibustion on spleen deficiency syndrome combined with gastric ulcer by observing the effects of moxibusting Zusanli, Zhongwan, Pishu, Weishu on the spleen deficiency syndrome, gastric tissues HE staining, Ulcer Index and expression of EGFR and phosphorylated ERK1/2proptein.Comparing the effect of moxibustion with classical prescription Sijunzi decoction on spleen deficiency syndrome combined with gastric ulcer, which can provide reliable experimental data for the clinical application of moxibustion.Methods:Fifty SD rats were randomly allocated to group A(blank group), group B(model group), group C(acupoints moxibustion group), group D(non-acupoints moxibustion group), group E(Sijunzi Decoction group). Rat of deficiency syndrome of the spleen model was made of the method of purgation with dahuang drugs of bitter flavour and cold nature except group A, and using the absolute alcohol fills method prepared rat model of spleen deficiency syndrome combined with gastric ulcer on this basis. Observed spleen deficieney symptom according to the Animal Model of Spleen Deficieney and the integral table of spleen deficieney symptoms.Used the method of Guth to calculate Ulcer Index.Histological changes in the gastric mucosa were observed by light microscopy.The contents of EGFR were detected by immunohistochemistry and the phosphorylation level of phosphorylated ERK1/2(p-ERK1/2) protein determined by western-blot.Results:1.In the process of creatiing spleen deficiency models, Model rats’defecate bagan to appear dilute and unformed with the prolonging of time,;They bagan to appear anorexia and reduced their food, mental state worse than before and the rate of stimulated defecation was increased;weight loss, became thin, back hair loose, fatigue, curl up together, which explain the model of spleen deficieney is successful.2.The symptoms of splenic of rats with spleen deficiency syndrome combined with gastric ulcer can improve by moxibusting on Zusanli, Zhongwan, Pishu, Weishu, compare with model group and non-acupoints moxibustion group before and after treatment, the difference was significant between them (P<0.01); The integral of acupoints moxibustion group were slightly lower than Sijunzi Decoction group after treatment, there was no statistically significant(P>0.05).3.Histological changes in the gastric mucosa can be improved by moxibusting on Zusanli, Zhongwan, Pishu, Weishu, compare with model group, non-acupoints moxibustion group and Sijunzi Decoction group, the cells of acupoints moxibustion group’gastric mucosal surface were more complete, the damage was significantly improved.4.The UI of rats with spleen deficiency syndrome combined with gastric ulcer can be reduce by moxibusting on Zusanli, Zhongwan, Pishu, Weishu, compare with the model group, the difference was statistically significant(P<0.01); although the UI of acupoints moxibustion group is lower than non-acupoints moxibustion group and Sijunzi Decoction group, there was no statistically significant(P>0.05).5.The EGFR of gastric tissue and the phosphorylation level of phosphorylated ERK1/2(p-ERKl/2) protein can be improve by moxibusting on Zusanli, Zhongwan, Pishu, Weishu, compare with the blank group, the model group, and the non-acupoints moxibustion group, the difference was significant between them (P <0.01); The EGFR of gastric tissue and the phosphorylation level of phosphorylated ERK1/2(p-ERKl/2) protein of acupoints moxibustion group is lower than Sijunzi Decoction group, there was no statistically significant(P>0.05).The lower Average gray value of EGFR in gastric tissue, the higher the positive reaction of EGFR and ERK1/2(p-ERK1/2) protein, which have significant correlation (r=-0.794, P<0.01)Conclusion:1.Moxibusted Zusanli, Zhongwan, Weishu and Pishu can obviously improve spleen deficiency syndrome of model rats, decrease the level of gastric mucosal injury and improve the histology of ulcerative injury of gastric mucosas, which are benifical for restoration of stomach function.2.Moxibusted Zusanli, Zhongwan, Weishu and Pishu can promote and improve the activation and expression of EGFR in gastric tissue.The activated EGFR mediated phosphorylation of ERK thus activating the EGFR/ERK signal transduction pathway, which can help resulting in cell proliferation and differentiation of a variety of biological effects and apoptosis, and reducing gastric mucosa damage and promoting the recovery of gastric mucosa Consequently.3.Moxibusting on Zusanli, Zhongwan, Pishu, Weishu, can improve the symptoms of splenic, promote restoratioin of gastric mucosa injury, the effect is just as Sijunzi Decoction group, and is better than non-acupoints moxibustion group, that’s suggest that Moxibustion is an effective way to treat spleen deficiency combined with gastric ulcer and is of certain point speciality, which reflects the"Comprehensive Effeet" consequence of Moxibustion-Acupoint-Meridian. |
