Based On The Theory Of Spleen And Tibetan God, The Mechanism Of Brain Intestinal Peptide In Rats With Spleen-deficiency Gastric Ulcer Was Studied

Objective:This study aimed to explore the relationship between spleen and stomach diseases and psychological and emotional disorders and its mechanism through both theoretical and experimental research,to reveal regulatory function of the spleen and stomach on emotions,and to provide experimental evidence for the theory of "spleen stores the spirit".Methods:Wistar rats were randomly divided into normal group,model group,Sijunzi Decoction group(model and Sijunzi Decoction administered by gavage),ranitidine group(model and ranitidine administered by gavage).Replication of spleen and stomach ulcer animal model was achieved through combination of purgative method with bitter-cold medicinals,irregular food intake with intragastric administration of vinegar.Then,the body weight,appearance,mental activity,food intake,defecation and other signs were observed and HE staining was performed.The expression of MTL?VIP?SS?EGF in gastric tissue was detected by immunohistochemistry and the expression of VIP and SS in serum was detected by enzyme-linked immunosorbent assay(ELISA).Results:1.In general,rats in the normal group were in good condition and their activities were normal.Rats in the model group had obvious weight loss,lassitude and abnormal fur,all of which were manifestation of spleen deficiency.Alleviation of symptoms of spleen deficiency with normal psychological and physical activity was observed in Sijunzi Decoction group.And symptoms and signs of rats in ranitidine group were similar to those of model group.2.The pathological changes of the gastric tissue were observed using HE staining under light microscope.The gastric mucosal lesion damage was obvious with disappearance of mucosal epithelium,exposure of the basal layer and hematopedesis in the model group.The morphological structure of gastric tissue in normal group and Sijunzi decoction group was normal.And inflammatory cell exudation of in submucosal layer was observed in ranitidine group.3.The expression of MTL?VIP?SS?EGF in the gastric mucosa of the rats was detected by immunohistochemistry.It showed that levels of MTL?VIP?SS?EGF in the model group were significantly lower than those in the normal group(P<0.05),while the expression of MTL?VIP?SS?EGF in the Sijunzi Decoction group and ranitidine group was significantly higher than that of model group(P<0.05).4.Detected by ELISA,the levels of VIP and SS in serum of model group were significantly lower than those of normal group(P<0.05).And the expression of VIP and SS in the Sijunzi Decoction group and ranitidine group was higher than that in normal group and the difference was statistically significant(P<0.05).Conclusion:1.Gastric ulcer is one of common psychosomatic disorders.Patients with it usually experience psychological and emotional disorders such as anxiety and depression,which may be related with spleen and stomach dysfunction,especially the dysfunction of "spleen stores the spirit".2.Due to the observation that serum VIP and SS decreased in rats suffered from spleen deficiency and stomach ulcer,the mechanism of mental and emotional disorders may due to dysfunction of brain-gut axis regulation on the body,which results from malfunction of spleen and stomach and abnormal brain-gut peptide secretion.Besides,the regulation of gastrointestinal system and the central nervous system by brain-gut peptide may be the material basis of "spleen stores the spirit".3.Sijunzi Decoction can effectively improve the symptoms of spleen deficiency in rats and prevent the occurrence of gastric ulcer.As the result showed that expression of VIP and SS in gastric mucosa and serum of Sijunzi Decoction group was higher than that of model group,it can be hypothesized that Sijunzi Decoction(with function of supplementing qi and fortifying the spleen)could keep brain-gut axis in stable state through increasing the expression of VIP and SS,which protect and enhance the function of neurons.4.In this study,we obtained the experimental evidence for the influence of spleen and stomach dysfunction on mind and emotions,It thus laid a foundation for further exploring the control approach of molecular signals related to "spleen stores the spirit" and revealing its modern biological mechanism.
