ObjectiveThrough this experiment, to observe the influence of electro-acupuncture and moxibustion joint used on behavior and hippocampalCAl regions cell period adjustive factor-cyclinE,p21/cip in Alzheimer’s disease model rats, explore the mechanism of electro-acupuncture and moxibustion improving the learning and memory ability and affecting neuronal apoptosis in AD model rats. Expect to further reveal the mechanism of action of Chinese acupuncture treatment for AD, and provide experimental basis for acupuncture preventing and treating AD better in clinical.MethodsAfter screened by the Y-type water maze(eliminated the rats whose react ion are too sensitive or too slow),wistar rats were randomly divided into four groups:normal group, sham-operation group, model group, electro-acupuncture and moxibustion group(hereinafter referred to as the treatme nt group),10rats per group. Aβ1-42was injected from the bilateral hip pocampal CA1regions by microinjector to produce AD rat models,"baih ui"(DU20) and "shenshu"(BL23) were stimulated by electro-acupunct ure and mild moxibustion in the treatment group after the modeling,and start the Morris water maze positioning navigation and space exploratio n experiments after treatment is completed, the experiment lasted for fiv e days, drawn at the end of the experiment the day. Immunohistochemi stry and Western blot were used to determine the content changes of cy clinE, p21/cip in rat hippocampi area, use SPSS Statistics17.0statistical software for data Statistical analysis.Results1. Behavior detection results:positioning navigation and space exploration experiments showed that the incubation period of escape, number of across the former platform and residence time had no significant difference between normal group and sham-operation group(P>0.05).Compared with the model group, the results of the normal group, sham-operation group and treatment group were significantly different (P<0.05),the behavior detection results of the treatment group was significantly better than the model group (P<0.05); Compared with the treatment group, the normal group and sham-operation group were significantly different (P<0.05).2. Immunohistochemistry results showed that the expression of cyclinE and ap21/cip had no significant difference beteen the normal group and sham-operation group(P>0.05); compared with model group, the expression of cyclinE and ap21/cip in the hippocampal region of the rat of the normal group, sham-operation group and treatment group were significantly different(P<0.05), which, the cyclinE positive cell number of the model rats was significantly more than the other three groups. And the p21/cip positive cell number was significantly less than the other three groups(P<0.05); compared with treatment group, the p21/cip positive cell number of the normal group and sham-operation group was significantly different(P<0.05).3. Western blot results showed:After two courses of treatment, compared with the normal group and sham-operation group,the expression of the cyclinE increased in rat hippocampi area on model group(P<0.05), compared with the model group,the treatment group decredsed obviously(P<0.05); and compared with the normal group and sham-operation group, the expression of the p21/cip decreased evidently on model group(P<0.05),compared with the model group,the treatment group incredsed evidently(P<0.05).Conclusion1. According to the fundamental pathogenesis of AD with deficiency of the kidney and encephalon and the spirit apraxia,we choose "Baihui" and "shenshu" to be stimulated by moxibustion and electro-acupuncture, that can play a therapeutic effect of nourishing kidney and filling encephalon, brighting brains and increasing intellect.2. The learning and memory function of AD model rats can be effectively improved by the united therapy of electro-acupuncture and moxibustion, alleviate the symptoms caused by the continuous degradation of memory and cognitive function, thus delaying the aging process.3. Electro-acupuncture and moxibustion can significantly reduce the over-expression of cyclinE in hippocampal CA1region of the AD model rats, increase the expression of p21/cip, regulate the cell division cycle, reduce neuronal apoptosis.4. The mechanism of acupuncture for the treatment of Alzheimer’s disease may achieved by regulating protein of the nerve cell cycle, of which positive control protein cyclinE and negative control protein p21/cip may play a key role.
