The Saxifraga genus (Saxifragaceae) consists of about400species, distribute throughout the world, there are about203species grown in China, mainly distribute in high cold areas such as southwest of China, Qinghai, Gansu Province. Saxifragaceae plants is used as traditional Chinese medicine and Tibetan medicine, which have the functions of detoxification and anti-inflammatory. Pharmacological studies have shown that plants of this genus has the functions of antibacterial and antiviral, antioxidant, anti-tumor, anti-male and female sex hormones, inducing fibroblast apoptosis, protect liver, etc. Plants of this genus contain a variety of flavonoids, terpenoids, essential oils, polyphenols, glycosides, organic acids and others. Saxifraga umbellulata is a common Tibetan, which has been used as a traditional folk medicine for the treatment of hepatitis cholecystitis, infuenza, fever, sore heat, and so on. This thesis deals with the investigation on the chemical constituents and antibacterial activity of Saxifrage umbellulata.The phytochemical investigation of the whole plants of Saxifrage umbellulata was studied. Using a variety of modern chromatographic separation methods (chromatography on silica gel, MCI column chromatography, chromatography, Sephadex LH-20gel column chromatography, preparative chromatography, etc.),27compounds were isolated and purified, the structure of25compounds were elucidated by spectroscopic identification, analysis (EI-MS, IR, UV, NMR etc.). These compounds belong to the flavonoids, coumarins, phenylalanine, terpenes, sterols and glycosides and others.The25compounds isolated from Saxifrage umbellulata were identified as quercetin(1), quercetin-3-O-β-D-glucopyranoside(2), Quercetin-3-O-β-D-Galactopyranoside(3), Quercet in-3-O-β-D-glucuronopyranoside-6"-butylester(4), Quercetin-3-O-p-D-[2"-(E)-feruloylgl ucopyranosyl]-7,4’-O-β-D-diglucopyranoside(5), Myricetin(6), syringetin(7),(3R)-8-Hyd roxy-6-methoxy-3-undecyl-3,4-dihydroisocoumarin(8),4-O-caffeoylqvinicacid(9),3,5-dica ffeoylquinic acid (10), Coumaricacid(11), Hirsutenone (12),(5R)-5-hydroxy-7-(3-methoxy-4-hydroxyphenyl)-1-(3,4-dihydroxyphenyl)-3-heptanone(13),(5S)-1,7-bis-(3,4-dihydroxyph enyl)-5-hydroxyheptane-3-one-5-O-β-D-Glucopyranoside(14),5(S)-butyloxy-1,7-di(3,4-d ihydroxyphenyl)-3-heptanone(15), loliolide(16),3,7,11,15-tetramethyl-2-hexadecen-1-ol (17), α-palmitoyl-β-linoleoyl-a’-linoleoyl-glycerol(18), Phytyl-1-hexcmoate(19), Pentad eanoic acid heptadecyl ester(20), DGDG(21),5α,8α-epidioxy-(22E-24R)-ergosta-6,22-di en-3β-ol(22), β-sitosterolpalmitare(23), β-daucosterol(24), Altertoxin1(25).In addition, the antibacterial activity of all the extracts were determined by Oxford cup method. The results showed that the extracts of acetic ether and n-butanol have fine antibacterial activity, but the extract of petroleum ether only show antibacterial activity to partial bacterium.The thesis reviews the chemical constitutents and pharmacological activity of the genus Saxifraga and describes the phytochemical research and antibacterial activity of Saxifrage umbellulala. This research provides a scientific basis for the further development and utilization of Saxifraga medicinal plant resources.
