Persimmon polysaccharide (PPS) is an important plant polysaccharides.A-bundant in China,Persimmon plants are rich in medicinal agents, chief among which is PPS. Hence extraction, separation, purification and structural analysis of PPS are important. Optimal conditions for said procedures are investigated in this paper.Crushed persimmon leaf tea extract is soaked in hot water to obtain a s-olution containing PPS which is then extracted via ethanol. Via single factor and orthogonal test setups we determined the effects of different variables on extraction yield. Starting with the most significant, variables are pH>Tempera-ture> Time> solid-liquid ratio. Optimalextraction parameters are pH=7,60℃,2h, and1:30ratio, resulting in an extraction yield of4.6%.After reconstitution in solvent,extracted PPS isdeproteinized via the Sevag method, bleached by hydrogen peroxide,and passed throughDEAE-52cellulose column chromatography to obtain four monomer fractions:PPS-1,thro ugh PPS-4,yield are5.1%,38.1%,11.2%,4.5%.IR and UV analysis suggest PPS is a glu-curonic acid containing glycoprotein.GC analysis of aldononitrile acetates deri-vatives of the four fractions showed they’re similar in composition,comprising of rhamnose, arabinose, xylose, glucose and galactose.HPLC of polysaccharide PMP derivatives showed relative compositionsof monosaccharides in each PPS fraction. Results are PPS-1:glucuronicacid:glucose:xylose:galactose:arabinos-e=0.41:1.49:5.23:1:0.56;PPS-2:rhamnose:glucuronicacid:galacturonic acid:gl-ucose:xylose:arabinose=0.14:0.21:0.22:0.19:0.68:1:0.47;PPS-3:rhamnose:gluc-uronic acid:galacturonic acid:glucose:xylose:arabinose=0.21:0.28:2.01:2,30:1:0.45;PPS-4:rhamnose:glucuronic acid:galacturonic acid:glucose:xylose:ara-binose=0.16:0.22:0.55:0.28:3.58:1:0.59.
