Objective:To research the effects of Dangua Recipe on glycolipid metabolism,insulin function,lesion of coronary artery,endothelial function of transgenic Apo E-/-mice which owning the feature of spontaneous atherosclerosis by using the synchronic treating phlegm and blood stasis.Thus revealing the therapeutic mechanism of improved endothelial function for curing diabetes with angiopathy, in order to provide experimental basis for clinical prescription in preventing and threating diabetic vascular lesions.Methods:1. The establishment of the diabetic model miceUsing streptozotocin peritoneally injected to ApoE-/-mouse,and then monitoring fasting blood glucose.Choosing32mice which fasting blood glucose (FBG)≥11. lmmol/1.2.groupingThe modeled mouses were stratified by body weight, and then, by blood glucose level from low to high, randomly divided into the model group,the DGR group,the combination group and and the pioglitazone group according to the random number table,8in each group. Another8euglycemia C57mouse of the same ages and strain were recruited as the control group.3. Processing methodThe interventions lasted for12weeks by gastrogavage.Monitored FBG, body weight, appetite, water intake, skin temperature, determination of glycosylated hemoglobin (HbAlc), triglycerides (TC), Low Density Lipoprotein-cholesterol (LDL-c).Fasting insulin, fasting c-peptide, endothelin-1(ET-1) were measured by radioimmunoassay, Observed coronary heart under light microscope.Assayed nitric oxide (NO)by nitrate reductase method. The kidney tissue VCAM-1. MCP-1level was analyzed with ELISA. The expression of VCAM-1、MCP-1mRNA in kidney tissue was detected with Real-time Quantitative PCR Detecting System.4. The data for statistical analysis.Results:1. Monitoring appetite and water intake showed:Appetite of Apoe-/-groups were less than C57group(P<0.01),in pioglitazone group was less than in DGR group(P<0.01).Water intake in model group, combination group, DGR group more than C57group(P<0.01or0.05)in pioglitazone group was less than in DGR group (P<0.05)2.Monitoring body weight showed:The body weight in the C57group was heavier than others all along (P<0.01OR P<0.05).The weight and its increment in the DRG group was eventually lighter than pioglitazone group.3.Monitoring skin temperature showed:the skin temperature of model group was higher than C57group(P<0.05),DGR group、combination group、pioglitazone group were higher than C57group(P<0.05),and DGR group was higher than model group (P<0.01)4Detecting FBG, HbA1C showed:FBG of DGR group and combination group as well as C57group but they were lower than model group(P<0.01);HbA1C of model group was higher than C57group(P<0.01),and the other Apoe-/-groups all lower than model group.(P<0.01)5. Detecting TC、LDL-c showed:all of Apoe-/-groups were higher than C57group(P<0.01),only DGR groups were both decreased(P<0.05).TC LDL-c of pioglitazone group were higher than DGR (P<0.01OR P<0.05)6. Detecting Fins、CP、FIRI showed:FIns、CP、FIRI in model group were increased.(P<0.01OR P<0.05) but in another group were no statistical significance (P>0.05)7.Pathomorphological changes:in model group,the tunica intima was obviously thickened,endothelial cells were degeneration and punctiform necrosis,the morphology of intima contained foam cells;but in DGR group endothelial cells only integrant degeneration,intima did not contain any foam cells and internal elastic lamina was clear.8. Detecting NO、ET-1showed:NO of all the groups were no statistical significance (P=0.517).ET-1of model group was higher than C57group (P<0.01),all of the other groups were lower than model group (P<0.01)9.Detecting VCAM-1、MCP-1by ELISA showed:VCAM-1、MCP-1of DGR groups were lower than model group (P<0.01),MCP-1of1of pioglitazone group was higher than DGR group (P<0.01)10.Detecting VCAM-l-mRNA、MCP-1-mRNA by qPCR showed:VCAM-1-mRNA、 MCP-1-mRNA of DGR groups were lower than model group (P<0.01) and in pioglitazone group were higher than DGR group (P<0.05)Conclusion:Dangua Recipe which the synchronic treating phlegm and blood stasis played a effective role in preventing endothelial function with diabetic angiopathy by comprehensively regulating glycolipid metabolism,relieving insulin resistance,protecting the vascular morphology.
