Ampelopsis grossedentata (Hand-Mazz) W.T.Wang is liana of Ampelopsis Michx. It can be used for dearing away heat and toxin material and inducing diuresis and reducing edema. It is used for the treatment of sore throat, red eyes and throat, icteric model hepatitis, etc. This study in order to comprehensively evaluate the quality of Ampelopsis grossedentata and screen the excellent clone,we determinate the content of effective componets of their different clones and medicinal parts.Besides,research their antioxidant antivity and the anti-inflammatory analgesic to understand the relationship between the difference of content of effective components and related efficacy. This thesis mainly is divided into six parts; the main contents are as follows:1. To optimize extraction process of total flavonoids, dihydromyricetin. myricitrin and myricetin from Ampelopsis grossedentata with ultrasonic wave by central composite design and response surface methodology. Ultimately determine the optimum process was30times the amount of65%ethanol ultrasonic extraction30min.2. Determined contents of total flavonoids, total phenols and polysaccharide in different clones and seasons Ampelopsis grossedentata by ultraviolet spectrophotometry, and Determinaed contents of dihydromyricetin.myricitrin and myricetin in theirs by HPLC method.to establish multiple index quality analysis system. The results showed that there was a large difference in different clones of Ampelopsis grossedentata, content of index in leaves was far greater than the content of the stems. In combination with principal component cluster analysis result,12different clones of Ampelopsis grossedentata divided into three categories. Assay results in different seasons of each index showed that Ampelopsis grossedentata clones’ content of each index change trend was consistent, in addition to the polysaccharide,content of each index was higher in spring and summer than in autumn.So picking Ampelopsis grossedentata should be more appropriate in4to6month.3. The common mode of the HPLC chromatographic fingerprints was established. There were13common peaks. Principal component analysis of its peak area, the three principal components extracted. Based on the clustering of analysis results of the three principal components’composite score.12different clones of Ampelopsis grossedentata divided into three categories. It consistented with the previous six indicators by clustering results. 4. Using GC-MS technique analyse the chemical consitituents of volatile oil from leaves and stems of Ampelopsis grossedentata. Forty kinds of compounds were identified from the leaves, accounting for62.64%of the total essential oil. Twenty-six kinds of compounds were identified from the leaves, accounting for50.64%of the total essential oil.And there are11compounds between leaves and stems. It preliminarily confirmed the aroma components of leaves and stems. And analyzed the differences in aroma components in their leaves and stems,it provided the basis for improving the taste of Ampelopsis grossedentata’s product--vine tea, and provides experiment basis for the comprehensive development and utilization of Ampelopsis grossedentata’s resources.5. The models of DPPH and ABTS were used to evaluate the antioxidant activity of the decoction from leaves and stems in different clones of Ampelopsis grossedentata, dihydromyricetin, and myricitrin and myricetin.The results showed that they had strong antioxidant activity, but there were some differences, and the antioxidant activity of leaves was stronger than stems. Different clones of Ampelopsis grossedentata’half clearance (IC50) and the contents of total flavonoids, total phenols, dihydromyricetin, myricitrin, myricetin were highly significant negative correlation by the analysis results of the correlation,.especially with total flavonoids.6. To study the activity of different levels of Ampelopsis grossedentata on the models of the swelling of mouse ear and acetic acid writhing in mice.The results showed the decoction of Ampelopsis grossedentata had certain anti-inflammatory analegesic activity, which grade "good"(AG-5) anti-inflammatory activity of the best, grade " medium "(AG-3) best analgesic activity. There were differences between different clones and dose of anti-inflammatory and analgesic activity and should be controlled in accordance with a reasonable amount of medicinal ingredient index content size.
