Ampelopsis grossedentata is a wild vine of the genus Vibrio,which has a flavonoid content of up to 45.5%,while dihydromyricetin is the most important flavonoid compound with a content of up to 39.5%,which is rare in the plant kingdom.Modern pharmacological studies have shown that Ampelopsis grossedentata has many effects such as anti-oxidation,hypoglycemic,blood pressure lowering,anti-inflammatory and analgesic,and has great application value in the fields of food and medicine.At present,the DMY content,extraction and purification and pharmacological effects have been studied,but its metabolic mechanism is still unclear.Therefore,this paper mainly tracks the dynamics of DMY content with PAL and CHI activity,and then the effects of two enzymes on the metabolic accumulation of DMY and the content of DMY were determined.In the aspect of rational development and application of the hypoglycemic activity of Ampelopsis grossedentata,this paper mainly discussed the flavonoids,flavones and their compounds in Ampelopsis grossedentata,the?-glucosidase and DPPH·scavenging model were used to analyze the effect of the nine tea-producing plants.The study was more suitabal for developing the plants with hypoglycemic products.The findings are as follows:1.The optimum conditions for PAL activity determination were:the optimum enzyme amount was 50?L,the reaction temperature was 55°C,the time was 30 min,and substrate concentration was 0.05 mol/L.The optimal conditions for the determination of CHI activity were:optimum enzyme amount 0.5 mL,reaction temperature 50?,time 30 min and substrate concentration 0.01 mol/L.2.The change of DMY content did not show significant regularity,in which the high content of plants in July was more,accounting for 54.55%.The PAL activity showed decreased gradually,and the enzyme activity in April was the highest,accounting for 48.48%.The CHI activity increased by 87.88%in June,which the activity of CHI increased by“W”pattern.The content of DMY in each producing area was positively correlated with PAL activity,and decreased with the growth of plants,and the showed a moderate positive correlation as a whole.The DMY content was negatively correlated with the of CHI viability as a whole.3.The ethanolic extract and the flavonoids from Ampelopsis grossedentata could inhibit?-glucosidaseto different extent,and the flavonol was obviously stronger than dihydroflavonol.The extracts from the leaves of Ampelopsis grossedentata from different flavones all had some effect on the inhibitory activity of?-glucosidase and DPPH·clearing ability.The content of dihydromyricetin and myricetin were positively correlated with?-glucosidase IC50?p<0.01?.The content of myricetin was significantly negative correlation and?-glucosidase IC50?p<0.05?.The effects of the three flavonoids components on DPPH·clearing ability were not significant.Inhibition of?-glucosidase and DPPH·scavenging ability of compound prescription were lower than that of sample.4.The contents of polyphenols and flavonoids in the water extracts of ten tea-producing plants were different,and the non-acidic components and acidic components had different inhibitory effect?-glucosidase and DPPH·clearing ability.The inhibition of?-glucosidase by non-acidic components was significantly different,and the values of IC50 was quite different.And none of them showed DPPH·scavenging effect.The inhibitory activity of acidic components on?-glucosidase was significantly stronger than that of the non-acidic components,and all of them had different DPPH·clearing ability.The activity of eagle tea and rattan tea was the strongest,but the extraction rate of eagle tea was significantly lower than that of rattan tea.Shiya tea has strong hypoglycemic activity in acid components.The acitivity of lobular Kudingcha was the weakest.The results show that rattan tea were more suitable to develop hypoglycemic products.
