This research aims to expound the processing principle of Gardenia jasminoides fried into carbon by studying on the chemical compositions of Gardenia, the processing technique, the content comparison of the volatile oil, the diterpenoid pigments and the iridoids systemic. Details are as follows:Chapter1:Summary on the Literatures of Gardenia jasminoides fried into carbonBy consulting and sorting the relevant literatures in the last50years, it has been summarized the processing history, the chemical compositions and the pharmacological actions of Gardenia jasminoids fried into carbon systematically, which promises this dissertation to be finished smoothly.Chapter2:Collection and preparation of gardenia sample piecesGardenia jasminoides which was recorded by pharmacopoeia of China is adopted as the raw herbal materials, which was prepared in raw Gardenia, Gardenia fried into carbon and several batches of commercially available Pieces, the experimental samples, which provide experimental sample for the research on Basic chemical substance and Comparison of pharmacological activities between raw gardenia and gardenia fried into carbon,take a good representation.Chapter3:Comparative study between gardenia and gardenia fried into carbon of HPLC fingerprint and identification of the major peaksA HPLC fingerprint analysis method, which is detected under two wavelengths(238nm,440nm), has been established for raw gardenia and gardenia fried into carbon.Through a comparative analysis of12batches of raw gardenia and15batches of gardenia fried into carbon. The HPLC fingerprint of gardenia was set up. Under238nm,nine peaks were identified in raw gardenia HPLC fingerprint and five peaks were identified in gardenia fried into carbon, All of them are iridoid glycosides.Under440nm, four peaks were identified in raw gardenia HPLC fingerprint and three peaks were identified in gardenia fried into carbon, which all are diterpene pigments ingredients. By using LC-MS, the structure of the main compound got further confirmation. Basically clarified chemical composition of the raw gardenia, gardenia fried into carbon and provided reliable clues for further quantitative principal component analysis of gardenia and gardenia fried into carbon.Chapter4:LC-MS Identification of Gardenia PiecesLiquid chromatography-electrospray ionization mass spectrometry method was used to analysize of the connotation of the material before and after gardenia Pieces of fried carbon change, the paper deduced the24compounds, which identified21compounds,3unknown compounds, the process of analyzing the trends and its fragments generated. The research can provide a reference for making the processing standard of gardenia jasminoides.Chapter8:The heating simulation validation experiments on variation of main components of Gardenia jasminoides before and after charring。In this research, G-1, G-2, G-3and C-1are mixed as a sample to have a heating simulation validation experiments. Through four ingredients trends observed during heating,the principle of variation of four main components was revealed. With the increasing of the heating temperature, four components occur the process of destruction or conversion, the iridoid glycosides are more Stable than pigments, C-1in the heating process, the gradual destruction, degradation and transformation process took place gradually transformed to C-2, C-3, C and other ingredients, with the severity of the destruction process, the C-1is the only remaining trace of the destruction of chemical ingredients.Chapter9:A Comparative research on the pharmacological effects of the group of main component in gardenia before and after charringPharmacological Experimental results show that gardenia has better antipyretic, anti-inflammatory effects of carbon after charring, gardenia fried into carbon can significantly shorten the prothrombin time, increased thrombus length, initially confirmed the gardenia fried into carbon not only enhance the role of the hemostatic, but also has similar antipyretic, anti-inflammatory effects of the raw gardenia.Chapter10:Results and DiscussionA Systemic research was taken on the Literatures of Gardenia jasminoides fried into carbon and variation before and after charring. A complete fingerprint HPLC of gardenia was established and the major peaks are identified. The research take a comparison of the Contents of the main chemical compositions of Gardenia iridoid glycosides and diterpene in Gardenia jasminoides fried into carbon. By comparing the raw carbon adsorption and tannin content, The variation of the absorption capacity of carbon and the content of the tanning between the gardenia jasminioides and its processed products can be used as index of the research on the gardenia jasminoides and its processed products. Heating simulation verification experiment was also taken to support the analysis results and developed modern scientific data which can make processing technology specification. By the above study, the changes of chemical compositions between the raw and charred Gardenia Jasminoides were revealed initially. Moreover, the study provided scientific evidence to expound the principle of frying into carbon.
